Objective: The present study investigated the protective effects of a pulsed electromagnetic field (PEMF) in a rat model of diabetes by analyzing oxidative/nitrosative stress parameters. Methods: Rats were randomly divided into four groups of eight each: a control group, a sham PEMF group, a diabetes group, and a diabetes+PEMF group. Diabetes was induced in the sham PEMF, diabetes, and diabetes+PEMF groups by treatment with 50 mg/ kg streptozotocin (STZ). Rats in the sham PEMF group were treated identically, without running the instrument. Following the development of diabetes, rats in the diabetes+PEMF group were treated with PEMF for 60 min/day for 4 weeks. Results: Levels of oxidants, such as malondialdehyde (MDA), nitric oxide (NO), and myeloperoxidase (MPO), and antioxidants, such as superoxide dismutase (SOD), glutathione (GSH), and catalase (CAT), were measured in blood and liver tissue samples. MPO, MDA, and NO levels were significantly higher, and SOD levels significantly lower, in the sham PEMF and diabetes groups than in the control group (p<0.05 each), whereas the levels of all these four (i.e.: MPO, MDA, NO, and SOD) in the diabetes+PEMF group were close to those in the control group. GSH levels were significantly lower in the sham PEMF, diabetes, and diabetes+PEMF groups than in the control group (p<0.05 each), whereas CAT levels were similar in all the four groups. Conclusion: Results indicate that PEMF affects MDA, NO, MPO, SOD, and GSH levels and regulates diabetes-associated damage by reducing oxidative stress and increasing the levels of antioxidants. PEMF may be a non-invasive treatment option for diabetes and associated complications.

Abstract. Oxidative damage is considered to play an important role in the pathogenesis of several diseases, such as diabetes mellitus (DM), atherosclerosis, cardiovascular complications and chronic renal failure. DM is associated with the oxidative stress and formation of advanced glycation end products (AGEs). Different drugs inhibit oxidative stress and formation of advanced glycation end products. Aminoguanidine (AG) has been proposed as a drug of potential benefit in prophylaxis of the complications of DM. Recent reports show a pro-oxidant activity of AG. Therefore we examined the effect of structural analogue of AG, its Schiff base with pyridoxal -pyridoxylidene aminoguanidine (PAG) on the level of selected markers of oxidative stress. We found that PAG decreased total damage to DNA in controls as well as in diabetic group of rats. However, we also found that PAG supplementation increases susceptibility of lipoproteins to oxidation and formation of conjugated dienes in both, diabetic as well as control animals. Its administration to diabetic rats decreases antioxidant capacity of plasma. Therefore, it is necessary to search for other structural modifications of AG that would combine its higher anti-diabetic activity with less toxicity.

Context: Chard is used as an antidiabetic agent by the diabetic patients in Turkey. Objective: The effect of chard extract [Beta vulgaris L. var. cicla (Chenopodiaceae)] on the antioxidant system and the expression of surfactant-associated proteins (SP) in the lungs of hyperglycemic rats were examined. Materials and methods: Hyperglycemia was induced by a single dose of streptozotocin (60 mg/kg) provided intraperitoneally. Fourteen days after the rats were rendered hyperglycemic, the chard (2 g/kg/d), insulin (6 U/kg/d), and chard plus insulin (as mentioned above) were administered to rats for 45 d. On day 60, rats' lungs were removed. Oxidative stress parameters and SP expression were assayed. Results: The lungs of hyperglycemic rats were characterized by the induced lipid and protein oxidation, elevated myeloperoxidase and xanthine oxidase activities, decreased glutathione levels, and reduced tissue factor and antioxidant enzymes activities (catalase, superoxide dismutase, glutathione peroxidase, and glutathione-S-transferase). Chard treatment alone and chard treatment combined with insulin were capable of achieving a regression of pulmonary oxidative stress, by inhibiting lipid and protein oxidation, and restoring the antioxidant system of hyperglycemic rats. SP-A expressions were significantly unchanged in all groups, whereas pro-SP-C and SP-D expressions were reduced in hyperglycemic rats. Pro-SP-C and SP-D levels were increased by chard and insulin administrations alone and combined in hyperglycemic rats. Discussion and conclusion: All treatments have a positive effect on the surfactant and antioxidant systems of the lungs of hyperglycemic rats. The best therapeutic effect was provided by treatment with chard extract alone in the compensation of hyperglycemic symptoms.
