Acute Effects of Ketamine on Memory Systems and Psychotic Symptoms in Healthy Volunteers

Abstract: N-methyl-D-aspartate (NMDA) receptor antagonists have been demonstrated to induce schizophrenia-like symptoms and cognitive impairment in humans. The NMDA receptor has been strongly implicated in memory, but research to date on the effects of NMDA antagonists has examined only some aspects of human memory functions. This study used a double-blind, placebo-controlled, independent groups design with 54 healthy volunteers to examine the effects of infusions of two doses (0.4, 0.8 mg/kg) of the NMDA antagonist ket… Show more

“…[45][46][47][48][49] Interestingly, increased response to acute AMPH treatment has been directly implicated in the positive symptoms of schizophrenia, 47 whereas acute pharmacological treatment with NMDA receptor antagonists may additionally induce memory impairments related to the cognitive symptoms of schizophrenia. 50, 51 Prenatal immune activation by the inflammatory agent PolyI:C in mice thus precipitates a wide spectrum of adult behavioral and pharmacological abnormalities related especially to the symptom profiles in schizophrenia.…”

“…7 A research line that is largely ignored in Dalenberg et al's review is the psychopharmacology of dissociative symptoms. Some drugs, notably ketamine, have been shown to produce severe dissociative symptoms along with memory dysfunctions in healthy volunteers (e.g., Morgan, Mofeez, Brandner, Bromley, & Curran, 2004). We consider this type of study important insofar as it may shed light on the issue of whether dissociative reactions may occur in the absence of a history of trauma.…”

“…Human evidence indicates that pharmacologic blockade of NMDA receptors impairs learning and memory (Morgan et al, 2004;Rockstroh et al, 1996), whereas post-learning administration of an NMDA agonist facilitates consolidation of fearful memories (Kalisch et al, 2009). …”
