Adhesion-based simple capture and recovery of circulating tumor cells using a blood-compatible and thermo-responsive polymer-coated substrate

Abstract: Circulating tumor cells (CTCs) have been a focus of study for metastatic cancer diagnostics, in in vitro anti-cancer drug screening to decide the chemotherapeutic course, and cancer biology research.

“…Thin films of the polymers were prepared by spin-coating 0.2 wt/vol % methanol solutions twice onto polyethylene terephthalate (PET) substrates. The polymer film is retained due to the van der Waals interaction between polymer and substrate, and the thickness is known to be around 100 nm by transmission electron microscopy . The films were placed in PBS and observed by AFM using a Cypher (Oxford Inst.…”

“…We have reported by TEM observation that the thickness of the PMEA film in the present conditions was around 100 nm. 78 Hence, the structure appeared only at the top layer region of the PMEA film. The protrusions were stable in water at least one 1 day.…”

“…The intermediate water is also observed in biocompatible/inert/non-fouling synthetic polymers ( Figure 1 d) [ 83 , 84 , 85 ]. The intermediate water dictates the cells behavior such as cellular attachment, proliferation, migration, and differentiation [ 86 , 87 , 88 ], and can be considered as a potential parameter for designing biomaterial scaffolds.…”
