Albumin and interferon-β fusion protein serves as an effective vaccine adjuvant to enhance antigen-specific CD8+ T cell-mediated antitumor immunity

Abstract: BackgroundType I interferons (IFN) promote dendritic cells maturation and subsequently enhance generation of antigen-specific CD8 +T cell for the control of tumor. Using type I interferons as an adjuvant to vaccination could prove to be a potent strategy. However, type I interferons have a short half-life. Albumin linked to a protein will prolong the half-life of the linked protein.MethodsIn this study, we explored the fusion of albumin to IFNβ (Alb-IFNβ) for its functional activity both in vitro and in vivo. … Show more

“…The postoperative decrease in serum Alb was an independent risk factor for postoperative complications [34]. Nutritional status is an important determinant of the immune response, and decreased Alb levels may be associated with an impaired antitumor immune response [35]. Therefore, judicious perioperative uid therapy may improve outcomes after surgery, especially in major gastrointestinal operated [36].…”

“…Generation of mouse pcDNA3-Alb-IFNβ Mouse IFNβ plasmid was performed as previously described. 20 We transfected Alb-IFNβ plasmid in Expi293F expression system kit (Thermo Fisher Scientific) and purified the proteins by the HiTrap Albumin column (GE Healthcare Life Sciences).…”

“… 17–19 Recently, we have improved on the drug kinetics and functionality of a type I interferon, interferon beta (IFNβ), by fusing it to albumin protein, creating Albumin-IFNβ (Alb-IFNβ). 20 This increases the half-life of IFNβ, and therefore enhanced cross-presentation by APCs and generated stronger antigen-specific T cell and B cell responses.…”

“…Lastly, the addition of albumin to IFNβ adjuvant expands its short half-life, and its co-administration with OVA or HPV E7 long peptides heightens the specific immunity of CD8 +T cells. A profound anti-tumor effect was observed in a TC-1 tumor model [ 72 ].…”
