Albuminuria is associated with too few glomeruli and too much testosterone

Abstract: Normally, the glomerular filtration barrier almost completely excludes circulating albumin from entering the urine. Genetic variation and both pre- and postnatal environmental factors may affect albuminuria in humans. Here we determine whether glomerular gene expression in mouse strains with naturally occurring variations in albuminuria would allow identification of proteins deregulated in relatively ‘leaky' glomeruli. Albuminuria increased in female B6 to male B6 to female FVB/N to male FVB/N mice, whereas th… Show more

“…C57BL/6 nephrin Y3F/Y3F mice developed a milder and later onset phenotype than CD-1 nephrin Y3F/Y3F mice, and the phenotype was more pronounced in male versus female mice, both of which are consistent with several reports describing strain-and sex-specific differences in the severity of renal disease in genetically modified mice. [24][25][26][27][28][29][30] C57BL/6 nephrin Y3F/Y3F mice showed significant albuminuria at .6 months of age ( Figure 5A). …”

“…46 Western blotting was performed on renal cortical tissue for Angpt1, Angpt2, VEGF-A, eNOS, VEGFR2, VEGFR2-P (Tyr 951 ), Tie-2 (C-terminus), Tie-2-P (Tyr 1100 ), antiphospho eNOS Ser 1177 , and total and phosphorylated nephrin. Tie-2 receptor phosphorylation was also assessed using immunoprecipitation as described.…”

“…In overtly healthy human populations, albuminuria varies with racial background 21,22 and sex. 23,24 This is recapitulated in inbred mouse strains: for example, young adult FVB mice have 8-fold greater albuminuria than B6 mice 25 and within each of these strains, males have 2-fold greater albuminuria than females. 25 The albuminuria range between female B6 and male FVB mice corresponds, after factoring for weight, to human albuminuria of 30-300 mg/day.…”

Notably, there are no significant differences in systemic blood pressure between FVB and B6 mice but the former have significantly fewer glomeruli/kidney.…”
