Abstract: Four lambda clones (lambda G11, lambda C4, lambda G14 and lambda G17) from the porcine MHC class III region were labelled with either biotin-14-dATP or digoxigenin-11-dUTP and hybridized in two different combinations to DNA fibres. The latter were prepared from agarose-embedded porcine peripheral lymphocytes lysed with proteinase K, then spread and fixed on poly-L-lysine-coated slides. Hybridization signals thus obtained confirm the order of the clones previously reported using pulsed-field gel electrophoresis… Show more

“…In this study, 2.7470.023 was adopted for pK f1 , which was estimated based on potentiometric titration (Sjo¨berg, 1998). Meanwhile, since the used FITEQL cannot fit the bulk deposition observed in the acidic supernatant during the hydroxide titration, a supposed reaction to simulate the interactions between aluminum species and silicic acid during the hydroxide titration was further introduced, where K f2 represents the formation constant for the produced complex.…”

“…Peripheral blood from a male horse (Reg #1219, Texas A&M University) with known fertility was used for (i) standard short-term pokeweed (Sigma) stimulated lymphocyte cultures to obtain interphase and metaphase chromosome preparations and (ii) agarose-embedded DNA plugs to obtain DNA fibers (31,32). DNA from individual BAC clones was labeled with biotin-16-dUTP and͞or digoxigenin-11-dUTP by nick translation using Biotin-or DIG-Nick Translation Mix (Roche Molecular Biochemicals).…”

“…DNA from individual BAC clones was labeled with biotin-16-dUTP and͞or digoxigenin-11-dUTP by nick translation using Biotin-or DIG-Nick Translation Mix (Roche Molecular Biochemicals). Hybridization to metaphase chromosomes or mechanically stretched DNA fibers was performed as described elsewhere in detail by us (30,(32)(33)(34). A minimum of 30 metaphase spreads and Ϸ50 interphase cells or DNA fiber hybridizations were captured and analyzed for each experiment by using a Zeiss Axioplan2 fluorescent microscope equipped with CYTOVISION͞GENUS application software, Version 2.7 (Applied Imaging, Santa Clara, CA).…”

“…[4,6,[18][19][20]) has received much less attention compared to that directed at their influence on growth (e.g., Refs. [2,3,5,8,[15][16][17][21][22][23]).…”
