Altered Ca2+ homeostasis in polymorphonuclear leukocytes from chronic myeloid leukaemia patients

Abstract: Background: In polymorphonuclear leukocytes (PMNL), mobilization of calcium ions is one of the early events triggered by binding of chemoattractant to its receptors. Besides chemotaxis, a variety of other functional responses are dependent on calcium ion mobilization. PMNL from chronic myeloid leukaemia (CML) patients that were morphologically indistinguishable from normal PMNL were found to be defective in various functions stimulated by a chemoattractant -fMLP. To study the mechanism underlying defective fun… Show more

“…Standard functional profiling reveals several GO term significantly overrepresented. It is widely known that the processes of Ca ++ homeostasis are often disordered in many cancer types ( 32 ). Therefore, the presence of GO term ‘ calcium-ion binding ’ among top enriched GO terms is of particular interest.…”

“…Such low [Ca 2+ ] i condition in FA cells, while peculiar per se, has so far been reported to our best knowledge only in chronic myeloid leukemia (CML) 21 22 and myeloid cells 23 , which is interesting in the perspective of FA cancer proneness and the high prevalence in FA of malignancies of myeloid origin 5 . The low [Ca 2+ ] i may operate as a protective mechanism in delaying both cell cycle and DNA replication, thus allowing more time for DNA repair.…”

“…Ряд даних свідчить про те, що проліферація клітин, зокрема злоякісний ріст, пов'язана з гі перкальціємією [13,15,16]. Навіть незначні ко ливання внутрішньоклітинної концентрації Са 2+ можуть призводити до активації проліферації чи загибелі клітин.…”
