Altered intracellular purine nucleotides in gamma‐irradiated red blood cell concentrates

Abstract: Gamma irradiation of SAGM-preserved RCCs leads to serious deterioration of the purine nucleotide metabolism of erythrocytes during storage, which can reduce the in vivo recovery of the transfused red cells.

“…The decreased concentration of extracellular Na + can be associated with water influx to the cytosol. This process and the absence of a selective mechanism of functionality, as performed in the spleen, could explain the increase of MCV in vitro with consecutive morphological RBC changes [5,14,15,16,17]. …”

“…The leakage of LDH to RBC medium can occur through direct membrane disruption (RBC lysis with concomitant release of LDH and Hb). An excessive LDH release at the end of the storage period could also be associated with depletion of intracellular energy-rich nucleosides (ATP, ADP, ITP (inositol triphosphate)) [37]. The enhanced energy loss that arises at the 2-to 4-week storage can lead to insufficient phosphorylation of membrane proteins and a mismatch of membrane skeleton assembly resulting in a serious reduction of the membrane's mechanical stability and in an increased permeability.…”

“…Before each measurement, the instrument was calibrated to a pH-value between 4AE0 and 7AE4 using standard controls. Intracellular nucleotides (ATP, ADP and AMP) were determined by highperformance liquid chromatography (HPLC) as described previously [13]. For sterility testing in all fractions, we used the BacT ⁄ ALERT Ò FA system for aerobic and anaerobic culture (Biomerieux, INC., Durham, Brasil S.A.) Statistics Statistical analyses were performed by Friedman's analysis of variance (ANOVA) test.…”
