An AFLP‐based linkage map of Zoysiagrass (Zoysia japonica)

Abstract: To construct an amplified‐fragment length polymorphism (AFLP)‐based molecular linkage map of zoysiagrass, the selfed progenies of a clone consisting of 78 individuals were analysed using 471 AFLP markers derived from 126 PstI/MseI primer combinations. Of these markers, 364 were grouped into 26 linkage groups. The maps covered a total length of 932.5 cM, with an average spacing of 2.6 cM between markers. This information proves useful for gene targeting, quantitative trait loci mapping, and marker‐assisted sele… Show more

“…However, there was no positional trend in the segregation distortion within each linkage group. This result differed from that in a previous study, in which many Euphytica (2009) 170:327-338 329 of the distorted loci were clustered at the bottom of LG2 (Cai et al 2004). …”

“…A total of 8.7% (40 of 458 loci) of the SSR markers showed significant segregation distortion in this population (P \ 0.01), a result similar to that previously reported for a zoysiagrass S 2 generation (6%) (Cai et al 2004). Similar numbers of distorted loci were detected in each parent, 'Tawarayama Kita 1' (20 of 335 loci) and 'Muroran 2' (22 of 314 loci), with only two identical markers showing significantly distorted segregation.…”

“…More recently, high-density genetic linkage maps have been constructed for zoysiagrass using AFLP and SSR markers (Cai et al 2004, Li et al 2009). In zoysiagrass, morphological characteristics that can be visually assessed are important factors when determining turf quality, while QTLs for tolerance of environmental stressors such as salinity and freezing may be widely distributed among species.…”
