Japanese iris (Iris japonica) is a popular perennial ornamental that originated in China; it has a long display period and remains green outdoors throughout the year. winter dormancy characteristics contribute greatly to the evergreenness of herbaceous perennials. Thus, it is crucial to explore the mechanism of winter dormancy in this evergreen herbaceous perennial. Here, we used the hybrid RNA-seq strategy including single-molecule real-time (SMRT) and next-generation sequencing (NGS) technologies to generate large-scale Full-length transcripts to examine the shoot apical meristems of Japanese iris. A total of 10.57 Gb clean data for SMRT and over 142 Gb clean data for NGS were generated. Using hybrid error correction, 58,654 full-length transcripts were acquired and comprehensively analysed, and their expression levels were validated by real-time qPCR. This is the first full-length RNA-seq study in the Iris genus; our results provide a valuable resource and improve understanding of RNA processing in this genus, for which little genomic information is available as yet. In addition, our data will facilitate in-depth analyses of winter dormancy mechanisms in herbaceous perennials, especially evergreen monocotyledons.

Pollen characteristics are very important for Iris interspecific hybridisation. In this study, the pollen viability and male meiosis were studied in yellow-flowered Iris dichotoma (Y2), I. domestica (S3) and their hybrids F1, F2 and BC1 (BC1-Y and BC1-S). The BC1-Y hybrids showed higher pollen viability than that of F1, F2 and BC1-S hybrids, which were between I. dichotoma (26.1%) and I. domestica (35.1%). Two sterile hybrids, F2-1 and BC1-S-1, exhibited more meiotic abnormalities (57.3% and 58.7%) than other individuals. During the first meiotic division, a diffuse diplotene stage was observed for the first time in the genus Iris. The meiotic abnormalities included non-congressed chromosomes, chromosome bridges, lagging chromosomes, unequal division, abnormally oriented spindle fibres, nonsynchronous division and polyad, and resulted in reduced pollen fertility. The relatively high frequency of 2n pollen grains was found in hybrids of BC1-Y-2, BC1-Y-1, BC1-S-2, BC1-S-3 and BC1-S-4. Our research provides a new resource for meiotic behaviour and pollen fertility of the genus Iris.

Bearded iris (Iris ×hybrida Hort.) is a large horticultural hybrid complex in the Iris genus, and the lack of understanding about its inheritance laws has seriously hindered the breeding process. From parental bearded iris ‘Indian Chief’ and ‘Sugar Blues’, four hybrid populations—including F1, F2, BC1P1, and BC1P2—were generated through hybridization. Fifteen key phenotypic traits, including plant height (PH), scape height (SH), length of fall (LF), width of fall (WF), length of standard (LS), width of standard (WS), and so on, were measured, and several genetic parameters (e.g., trait variation, heritability, trait correlation, distribution of flower color) were analyzed. The variation of phenotypic traits indicated that the F1 generation could produce larger flowers and a greater number of blooming stems than other generations, whereas backcrossing was beneficial at producing more flowers on one scape in the offspring of ‘Indian Chief’ and ‘Sugar Blues’. WF had the greatest broad-sense heritability (73.91%) among the 15 phenotypic traits, whereas the broad-sense heritability of SH was the lowest (2.06%). The correlation between a vegetative trait (PH) and a reproductive trait (WS) provided a path to early selection of germplasm. Furthermore, four important floral traits (LF, WF, LS, and WS) also correlated significantly to each other, thus simplifying the selection of larger flowers. Genes regulating fuchsia flower color were dominant over those for bluish purple flowers. Genetic effects of flower color in recurrent parents could be reinforced by backcrossing, thereby providing a potential way to modify flower color through hybridization.
