An Efficient Method for Direct Shoot Regeneration from Leaf Explants of Solanum nigrum Induced by Thidiazuron

Abstract: Direct shoot regeneration was established from leaf, petiole and nodal explants from in vitro and field grown plants of Solanum nigrum on Murashige and Skoog (MS), and Gamborg's (B5) media containing different levels of plant growth regulator, Thidiazuron (TDZ). Comparison of the individual effect of basal media, levels of TDZ, explant type or source of explants on shoot regeneration was tested. Percent shoot regeneration from leaf explant was highest (86.66%) followed by in vitro source of explant (83.65%), B… Show more

“…The success of potato in vitro culture depends on the explant, culture medium, and combination and concentration of plant growth regulators (PGRs) in the culture medium ( Afrasiab, Rashid & Akram, 2017 ; AL-Hussaini, Yousif & AL-Ajeely, 2015 ; Kumlay, 2014 ; Mohapatra et al, 2018 ). Shoot culture is the basic technique of potato biotechnology.…”

“…One plausible explanation suggests that TDZ might facilitate the accumulation or synthesis of native plant growth hormones, contributing to its observed effects (Maity et al, 2016). The promotion of growth such as callus formation, shoot regeneration, and somatic embryo formation take place when applied at higher concentration and axillary proliferation at comparatively lower concentrations (Haddadi et al, 2013;Ugandhar & Venkateshwarlu, 2015;Afrasiab et al, 2017). It might be due to its similar biological activity like other N6-substituted cytokinins such as N-Ndiphenylurea and N-(2-chloro-4-pyridyl)-N′phenylurea.…”

“…However, 6-Benzylaminopurine (BAP) and thidiazoron (TDZ) are also used as plant hormones for in vitro regeneration (Ehsanpour and Jones, 2000). TDZ is a substituted Nphenyl urea which is an important regulator that promotes the maturation of somatic embryogenesis and increases the shoot induction of callus in many in vitro regeneration systems of plants (Afrasiab et al 2017). Na + /H + antiporter genes are important for improving the plant's tolerance to salinity using genetic engineering.…”
