An Immunosensor Using Electroactive COF as Signal Probe for Electrochemical Detection of Carcinoembryonic Antigen

Abstract: Two kinds of two-dimensional (2D) covalentorganic frameworks (COF) were used to construct a sandwichtype electrochemical immunosensor for a proof-of-concept study. Vinyl-functionalized COFTab-Dva could be linked with Ab1 by the thiol−ene "click" reaction. Electroactive COFTFPB-Thi was modified with gold nanoparticles (AuNPs) to ensure the successful connection with Ab2 through Au−S bond. Meanwhile, electroactive COFTFPB-Thi was used to as signal probe to realize both the detection of carcinoembryonic antigen (… Show more

“…Common tumor markers include carcinoembryonic antigen (CEA) and carbohydrate antigen (CA) 24-2 [ 16 , 17 ]. CEA is a structural protein and one of the components of cell membranes; it can be detected in the serum of patients with CRC [ 18 ]. Elevated CEA is common in gastrointestinal tumors, urinary tract tumors, lung cancer, breast cancer, and other diseases.…”

“…According to a previous report [ 36 ], the redox mechanism is speculated to be that the nitrogroup contained in the structure of furazolidone is reduced under the synergistic catalysis of Pt and rGO. With the increase in the scanning rate, the peak current density of furazolidone increased ( Figure 3 c), and a good linear relationship was presented ( Figure 3 d), indicating that the reaction process was a typical surface control process [ 37 , 38 ].…”

“…When CEA is captured by Ab1@COF Tab‐Dva , two units of electrons are transferred between COF TFPB‐Thi (reduction) and COF TFPB‐Thi (oxidation); therefore, the amount of CEA can be quantified based on the changes in the peak signal intensity of the COF TFPB‐Thi redox reaction. [ 73 ] These COF‐based electrochemical immunosensors show wide detection range, low detection limit, and practicality, indicating outstanding application prospects for determining biomarkers in clinical analysis.…”
