An improved method for rearing Halotydeus destructor (Acari: Penthaleidae) in the laboratory

Abstract: We developed an improved method for rearing Halotydeus destructor in the laboratory. Mite numbers increased rapidly over the summer of 1992-1993 when fed on vetch, Vicia sativa cv Blanehefleur; in contrast mite numbers were low and fell over the second half of the summer of 1990-91 when fed on subterranean clover, Trifolium subterraneum cv Junee. On V. sativa at a fluctuating temperature from 11 to 18°C 200 mites produced a mean of 1308 and 1455 progeny per transfer in 2 years, a 6-7-fold rate of increase per … Show more

“…epizootics can build up rapidly in two-spotted spider mite populations when humidity is high (Brandenburg & Kennedy, 1982;Klubertanz et al, 1991). Another fungus, Verticillium sp., which caused H. destructor mortality in the laboratory (Ridsdill-Smith & Gaull, 1995), was not found attacking mites in pastures. Predatory mites were seen infrequently at these sites and were not counted.…”

“…Halotydeus destructor is thought to have a slightly shorter generation time than the Penthaleus spp. (Narayan 1962;Ridsdill-Smith and Gaull 1995;Ridsdill-Smith and Annells 1997), while that of P. sp. x has not been examined.…”

“…Laboratory rearing methods have been successfully established for H. destructor, providing up to 20,000 mites at a time for screening experiments (Ridsdill-Smith and Gaull 1995;Thackray et al 1997). However, culturing techniques for the Penthaleus spp.…”

“…A single mite was placed in a treated vial along with a vetch (Vicia sativa cv Blanchefleur) leaf, which was used for food (Ridsdill-Smith & Gaull, 1995) and increased humidity. We exposed mites to this rate, 2× this rate and a series of dilutions to develop dose-response curves.…”
