An improved perfusion fixation method for the testis

Abstract: A reliable and uniform vascular perfusion fixation method for the testis has been developed by using an initial washout solution containing a vasodilator and an anticoagulant. This is followed by a brief fixation with a sodium phosphate buffered formaldehyde-glutaraldehyde solution of conventional strenght, and then a second more concentrated aldehyde fixative solution containing picric acid. The method takes into account some of the unique features of the vascular supply of the male genital tract for its favo… Show more

“…The sheep were perfused through the descending aorta with a mixture of polyvinylpyrolidinone, heparin, and procaine hydrochloride in saline to clear the blood from the vascular network, with the excess perfusate being drained from the inferior vena cava (Forssmann et al, 1977). Once the perfusate from the inferior vena cava was clear, four of the animals were perfused with an India ink/gelatin mixture (Miller and Jee, 1980) and the remaining six animals were perfused with an India ink/ formalin mixture (Hayat, 1981).…”

“…A canula of suitable size was inserted into the straight intratunical portion of the testicular artery and a rinsing fluid (for composition see WROBEL et al, 1978) was instilled at low pressure. One ovine testis was fixed with Bouin's fluid; all other testes were fixed with solution I and II of FORSSMANN et al (1977). Cube-shaped blocks with a square length of about 1 mm were cut from the tissue and postfixed in solution II.…”

“…Immediately after slaughter in Assiut (Egypt) abattoir, the testes were removed and fixed by vascular perfusion via the straight intratunical portion of the testicular artery. Following a rinsing procedure, the fixatives I and II of FORSSMANN et al (1977) were instilled at low, steady pressure (for details see WROBEL et al, 1978WROBEL et al, , 1988. Small blocks from seminiferous parenchyma were postfixed in solution II for 3-5h, washed in 0.2M phosphate buffer, osmicated (1% Os 04), dehydrated in a graded series of ethanol and embedded in ERL (SPURR, 1969).…”
