An improved sample preparation method for the sensitive detection of peptides by MALDI‐MS

Abstract: We describe here an optimization study of the sample preparation conditions for sensitive detection of peptides by matrix-assisted laser desorption/ionization mass spectrometry (MALDI-MS). Among many factors in the conditions, we varied the percent acetonitrile in the peptide solution, the percent acetonitrile in the matrix solution and the α-cyano-4-hydroxycinnamic acid (CHCA) concentration in the matrix solution. CHCA was chosen because it is the most frequently used matrix for analyzing peptides. The well-e… Show more

“…29 The method is simple in concept and design with limit of detection at femtomol levels. 30 The release kinetics of vancomycin upon 10 4 Staphylococcus aureus addition was also confirmed by a qualitative MALDI-ToF-MS analysis which is more reliable and sensitive compared to UV absorbance method. 29 Fig.…”

“…The synthesis and applications of the α-cyanocinnamic acid derivatives such as 4-chloro-α-cyanocinnamic acid (Cl-CCA) [5], ( E )-2-cyano-3-(naph-thalen-2-yl)acrylic acid (NpCCA) [6] and (2 E )-3-(anthracen-9-yl)-2-cyanoprop-2enoic acid (AnCCA) [6] have now been reported. Research studies indicate that 4CHCA is still important for peptide and protein analysis [79], and employing hybridization of matrix [10] or graphite supported preparation [11] makes it a more suitable matrix.…”

“…3 Among a variety of matrices used in MALDI-TOF MS for the analysis of biomolecules, 2,5-dihydroxybenzoic acid (2,5-DHB) is commonly used to study peptides and proteins, whereas 2,6dihydroxybenzoic acid (2,6-DHB) is used to study oligonucleotides and polysaccharides. 3,4 Trifluoroacetic acid (TFA) is commonly used as a matrix additive in MALDI-MS. 5 Phosphoric acid (PA) was recently introduced as a novel matrix additive for the analysis of phosphopeptides and nonphosphopeptides. 6,7 In this study, we compared the effectiveness of TFA and PA as matrix additives to identify ideal conditions for analyzing glycans released from ovalbumin.…”
