Inflammation is a local defensive reaction of a host to cellular injury or infection. Prolonged inflammation can contribute to pathogenesis of many disorders. Identification of naturally occurring phytoconstituents that can suppress inflammatory mediators can lead to the discovery of anti-inflammatory therapeutics. Acacia ferruginea is used traditionally to treat numerous ailments including hemorrhage, irritable bowel syndrome and leprosy. The present study evaluated the anti-inflammatory activity of A. ferruginea extract against acute (carrageenan) and chronic (formaldehyde) inflammation in Balb/c mice. Pre-treatment with A. ferruginea extract (10 mg/kg BW) for 5 consecutive days via intraperitonial (IP) administration significantly inhibited subsequent induction of paw edema in both models; the effects were comparable to that of the standard drug indomethacin. The results also showed the A. ferruginea extract significantly inhibited nitric oxide (NO) synthesis and iNOS expression (as measured in serum), diminished inflammation in -and neutrophil infiltration to -the paw tissues and led to a reduction in the number of COX-2 + immunoreative cells (as evidenced by histologic and immunohistochemical analyses) in the paws relative to those in paws of mice that received the irritants only. Further, in vitro studies showed the extract could significantly scavenge free radicals generated as in DPPH and NO radical generating assays. Taken together, the results showed that A. ferruginea extract imparted potent anti-oxidant and -inflammatory effects, in part by maintaining oxidative homeostasis, inhibiting NO synthesis and suppressing iNOS and COX-2 expression and so could potentially be exploited as a potential plant-based medication against inflammatory disorders.

Context: Red algae have been recognized as a rich natural source of compounds possessing interesting biological and pharmacological activities. Objective: This work investigates anti-inflammatory, analgesic and gastroprotective activities of MeOH/ CH 2 Cl 2 crude extract and its fractions F1 (50% MeOH) and F2 (80% MeOH) from the whole alga plant Laurencia obtusa Hudson (Rhodomelaceae). Materials and methods: Anti-inflammatory activity was evaluated in vitro using cytometric bead array (CBA) technology to follow up the secretion of tumour necrosis factor alpha (TNF-a) in lipopolysaccharide activated THP-1 monocytic cells at doses of 10-250 lg/mL and in vivo using carrageenan-induced paw oedema in Wistar rats at doses of 25, 50, 100 and 200 mg/kg. Crude extract and fractions were tested at the doses of 25, 50, 100 and 200 mg/kg for peripheral and central analgesic activity by acetic acid-induced writhing test and hot-plate method, respectively, in Swiss albino mice. Gastroprotective activity was evaluated using HCl/ethanol-induced gastric ulcer test in rats at doses of 25, 50, 100 and 200 mg/kg. Results: Crude extract, F1 and F2 showed an interesting inhibition of TNF-a secretion with IC 50 values of 25, 52 and 24 lg/mL, respectively, and a significant anti-inflammatory activity in vivo (p < 0.01), 3 h after carrageenan injection, the oedema inhibition was 55.37%, 52.18% and 62.86%, respectively, at the dose of 100 mg/kg. Furthermore, they showed a significant peripheral analgesic activity with 53.79%, 55.92% and 57.37% (p < 0.01) of writhing inhibition, respectively. However, no significant activity was found in the hotplate test. An interesting gastroprotective effect was observed with crude extract and its fractions F1 and F2 with a gastric ulcer inhibition of 65.48%, 77.42% and 81.29%, respectively, at the dose of 50 mg/kg. Discussion and conclusion: These results suggest that L. obtusa might be used as a potential source of natural anti-inflammatory and analgesic agents with gastroprotective effect. ARTICLE HISTORY

In this study, the analgesic activities and the mechanism of anti-inflammatory activities of 50% ethanol extract from TS (ETS) in vivo were investigated. Investigations were performed in mice with 2 analgesic models: acetic acid-induced writhing response and formalin-induced paw licking. The anti-inflammatory effect was tested by λ-carrageenan (Carr)-induced mice paw edema. These analgesic results indicated that ETS at a dose of 0.5 and 1.0 g/kg reduced the acetic acid-induced writhing responses and the licking time in the late phase of the formalin test. Moreover, Carr-induced paw edema was significantly reduced when ETS (0.5 and 1.0 g/kg) was administered 1 to 5 h after Carr injection. ETS reduced the level of malondialdehyde (MDA) in the edema paw by increasing the activities of antioxidant enzymes, such as superoxide dismutase (SOD), glutathione peroxidase (GPx), glutathione reductase (GRx), and catalase (CAT), in the liver and reducing tumor necrosis factor- (TNF-), interleukin (IL)-1, and IL-6 activities in the edema paw. This study demonstrates the analgesic and anti-inflammatory effects of ETS, thus verifying its popular use in traditional medicine.
