ÖZETİdrarda testosteron (T) ve epitestosteronun (E) tayini için, ince tabaka kromatografisi ile kombine basit ve duyarlı gaz kromatografik bir yöntem tanımlanmıştır. İdrardaki testosteron ve epitestosteron asit hidrolizi ve ekstraksiyondan sonra ince tabaka kromatografisi (İTK) ile ayrılmış ve elüe edilmiştir. Elüat asetillenmiş, tekrar İTK ile izole edilmiş ve iç standart ilave edilip gaz sıvı kromatografisinde (GSK) analiz edilmiştir. Bu yöntemle verim testosteron için % 73.79 ± 3.9 ve epitestosteron için % 75.94 ± 7.9, duyarlılık ise 0.05 µg olarak bulunmuştur. Geliştirilen yöntem normal erkek ve kadın idrarları ile ilaç verilen kişilerin idrarlarına uygulanarak T E oranlan saptanmış-tır. Normal erkek idrarlarında bu oran 1.96 ± 0.62, normal kadın idrarlarında 2.62 ± 0.32, ilaç verilen kişilerin idrarlarında ise 4.88 ± 1.78 olarak bulunmuştur. Bulgular doping kontrolü açısından değerlen-dirilmiştir. SUMMARYA simple and accurate gas chromatographic method for the determination of testosterone (T) and epitestosterone (E) in human urine is described. SummaryAdult male volunteers were administered orally 10 mg of the respective drug. Urine samples were investigated by microcolumn liquid chromatography and capillary gas chromatography combined with on-line mass spectrometry. The former technique was found to be usable for the detection of these drugs. The latter method appeared to be superior in terms of sensitivity and for metabolism studies. The major excretion products of dexamethasone were identified as isomeric 6-hydroxy metabolites. Dexamethasone as such and its 20-hydroxy metabolite were found in minor quantities. Unlike dexamethasone, triamcinolone is excreted largely unchanged from the human body. Two metabolites were identified as 11-keto and 4,5-dihydrotriamcinolone. With the gas chromatographymethods developed, abuseof these drugscan bedetected up to 24 h after administration of a 5 mg single dose.

Determination of methyltestosterone using flow injection with chemiluminescence detection

Methyltestosterone (MT) enhanced the chemiluminescence of potassium permanganate-sodium thiosulphate system in sulphuric acid medium, and this was used as the basis of a novel flow-injection chemiluminescence method for the determination of methyltesterone. Under optimum conditions, the linear response of MT was in the range 3.0 x 10(-8)-8.0 x 10(-6) g/mL, and the detection limit for determination of MT was 1.0 x 10(-8) g/mL. The method was applied to the analysis of fish with recovery in the range 88.0-113.8% with an average RSD of 1.66% (n = 8).
