Analysis of nickel species in cytosols of normal and malignant human colonic tissues using two dimensional liquid chromatography with ICP-sector field MS detection

Abstract: A method has been developed that allows the nickel species in healthy and neoplastic tissues from cancer subjects to be compared. It is based on the coupling of sequentially applied anion-exchange and size-exclusion chromatography using an ICP-sector field mass spectrometer as detector. The method allows the resolution of seven nickel species in tissue extracts, which can be considered as fingerprints of nickel speciation. A qualitative difference in Ni-binding biomolecules between cytosols of normal and malig… Show more

“…Exposure to 1 mM Cd triggered a transient growth arrest. Consequently, we determined, by Inductively Coupled Plasma Mass Spectroscopy (ICP-MS, [30] , [31] ), the amount of Cd that exponentially growing cells retain at 30 min, 120 min, 270 min and 24 hrs under our culture conditions (ASW-AA-Cyst medium) and showed that cells always retain several percents of the amount of Cd (1 mM) added to the culture ( Table S1 ). However, these percentages differ from one culture to another, except for the 24 hrs timepoint.…”

“…The solution was diluted 1000 times in HNO 3 2% prior analysis by ICP-MS. The washing supernatants were diluted 10 times in HNO 3 2% prior analysis by ICP-MS. Rhodium was used as internal standard and quantifications were performed by external calibration [30] , [31] (Agilent 7500 ICP-MS, Agilent Technologies, Tokyo, Japan).…”

“…199 Bidimensional AE-SEC-HR ICP MS allowed the resolution of 7 Ni species in tissues from cancer patients but no difference in binding patterns between cytosols of normal and malignant colon tissues could be revealed. 206 Reversed-phase HPLC-ICP MS was used for the analysis for metal-MT complexes in rat liver and kidney. 207 Capillary electrophoresis offers a better separation efficiency of MT isoforms and their complexes than HPLC.…”

“…SEC‐AE HPLC‐ICP MS and stable‐isotope labeling were used to study the Cu turnover in marine animals (Mason & Borja, 2002). Bidimensional AE‐SEC‐HR ICP MS allowed the resolution of seven Ni species in tissues from cancer patients, but no difference in binding patterns between cytosols of normal and malignant colon tissues could be revealed (Bouyssiere et al, 2004). Reversed‐phase HPLC‐ICP MS was used for the analysis for metal‐MT complexes in rat liver and kidney (Polec et al, 2002).…”
