Angiocrine Factors Modulate Tumor Proliferation and Motility through EphA2 Repression of Slit2 Tumor Suppressor Function in Endothelium

Abstract: It is well known that tumor-derived proangiogenic factors induce neovascularization to facilitate tumor growth and malignant progression. However, the concept of "angiocrine" signaling, in which signals produced by endothelial cells elicit tumor cell responses distinct from vessel function, has been proposed, yet remains underinvestigated. Here, we report that angiocrine factors secreted from endothelium regulate tumor growth and motility. We found that Slit2, which is negatively regulated by endothelial EphA2… Show more

“…Murine pulmonary microvascular endothelial cells (MPMECs) were isolated from 1-to 3-month-old Rictor fl/fl or Raptor fl/fl mice and maintained in EGM-2 medium (Lonza) for Ͻ5 passages, as described in our previous studies (22)(23)(24)(25). For adenovirus-mediated expression of Cre (Ad-Cre) or Ad-LacZ, MPMECs were seeded into 10-cm dishes at 70 to 80% confluence and infected with 10 7 PFU ml Ϫ1 virus for 48 h prior to biological assays and Western blot analysis.…”
