Angiography of Fluoresceinated Anti–Vascular Endothelial Growth Factor Antibody and Dextrans in Experimental Choroidal Neovascularization

Abstract: To determine if anti-vascular endothelial growth factor antibody and a range of dextrans with varying diffusion radii and molecular weights are permeable through experimental choroidal neovascularization (CNV). Methods: Choroidal neovascularization was induced in 10 cynomolgus monkey retinas by means of argon laser injury. Digital fundus fluorescein angiograms were performed with fluorescein sodium, fluoresceinated IgG antibodies (anti-vascular endothelial growth factor and a control antibody), and fluorescein… Show more

“…Direct experimental evidence that intravenously injected fluoresceinated bevacizumab or control IgG leaks from CNV in a primate model was then obtained. 84 However, the safety of prolonged systemic exposure to an anti-VEGF agent was a potential concern. Although toxicological studies of young adult monkeys receiving intravenously administered bevacizumab had not revealed any major safety signals, 85 the possibility that adverse effects might occur in an elderly population such as AMD patients, frequently affected by cardiovascular diseases, could not be ruled out.…”

The potentially better and longer visualization of this modified dye may be helpful for an improved detection and monitoring of pathological alterations in various eye diseases.…”
