Animal models of fulminant hepatic failure: A critical evaluation

Abstract: Few conditions in medicine are more dramatic or more devastating than acute liver failure. Our understanding and treatment of this condition have been limited by the lack of satisfactory animal models. The most widely used models consist of surgical anhepatic and devascularization procedures and hepatotoxins, such as galactosamine and acetaminophen. Potential disadvantages with surgical models are their inability to recreate the inflammatory milieu that exists in acute liver failure and their reliance on surgi… Show more

“…On the other hand, in living donor liver transplantation using extremely small-sized grafts, the transplanted graft could be supported by the remnant native liver [4,16]. As possible applications of the current APOLT model, several experiments could be designed using rats with chemically or surgically induced fulminant hepatitis [17], metabolically diseased rats [18], marginal donor grafts like fatty grafts [19]. Now we are investigating the use of warminjured grafts from non-heart-beating donors using the current APOLT model.…”

“…In the worldwide literature, the model for inducing acute liver failure via toxins in rats has been mostly used to test support therapies consist of d-galactosamine intraperitoneal administration, because it is potentially reversible, presenting non-significant extra-hepatic toxicity and has the best-defined hepatotoxic dosage 10 .…”
