Anti-Inflammatory Isoflavonoids from the Stems ofDerris scandens

Abstract: Fractionation of the aqueous extract of Derris scandens stems extract using tests for eicosanoid inhibition resulted in the isolation of three isoflavonoids, genistein, its 7- O-alpha-rhamno(1-->6)-beta-glucosyl glycoside, a new compound, and two known isoprenyl derivatives 3'-gamma,gamma-dimethylallylweighteone and scandenin. The isoprenylated compounds showed a high inhibitory effect on eicosanoid production in vitro but HPLC analysis showed that the genistein accounted for most of the activity of the total … Show more

“…The major active constituents of D scandens stem extracts are benzyls and isoflavones, including genistein, coumarins, scandinone, scandenin, prenylated isoflavones, and isoflavone (Rukachaisirikul et al, 2002;Laupattarakasem et al, 2004;Rao et al, 2007). D scandens stem extracts has been widely used in traditional medicine for the treatment of several diseases such as arthritis, muscular pain, and inflammation (Tiangburanatham, 1996).…”

“…Our study supports the hypothesis that the mechanism of analgesic action of D. scandens is likely to NSAIDs. The isoprenylated compounds, isolated from D. scandens stems, showed a high inhibitory effect on eicosanoid production for COX and 5-lipoxygenase [19]. Although the additivity analgesic interaction of the combination between the D. scandens extract drug and diclofenac could not significant reduce the dose of each drug alone, it may reduce incidences of side-effects.…”

“…and leukotriene-B4 (LTB4) generation in the same manner, similar AEs of both can be produced (Wongsinkongman et al, 2013;Kuptniratsaikul et al, 2011;Mahabusarakam et al, 2004;Laupattarakasem et al, 2004). In Thailand National List of Essential Medicines (NLEMs), D. scandens should be used with caution in peptic ulcer patients because it may irritate the gastrointestinal mucosa.…”

“…scandens extract exhibited good anti-inflammatory effect by inhibition of cyclooxygenase 1 (COX-1) and leukotriene-B4 (LTB4) generation, reduction of eicosanoid synthesis via both cyclooxygenase and lipoxygenase pathways and myeloperoxide release (Wongsinkongman et al, 2013;Kuptniratsaikul et al, 2011;Mahabusarakam et al, 2004;Laupattarakasem et al, 2004).…”
