Anti-inflammatory γ- and δ-tocotrienols improve cardiovascular, liver and metabolic function in diet-induced obese rats

Abstract: In rats, δ-tocotrienol improved inflammation, heart structure and function, and liver structure and function, while γ-tocotrienol produced more modest improvements, with minimal changes with α-tocotrienol and α-tocopherol. The most important mechanism of action is likely to be reduction in organ inflammation.

“…These protective effects are not shared with TF [101]. TRF, in particular its γand δ-isomers, reduced hypertension in rats with diet-induced metabolic syndrome (MetS) [57,102,103]. It improved vascular endothelial function by promoting vascular relaxation [104] and eNOS activity that enhances NO production, a potent vasodilator [105] in spontaneously hypertensive rats (SHRs), leading to a decrease in systolic BP.…”

“…The metabolic changes include the reduction of abdominal circumference, omental fat, FBG, systolic BP, TG, NEFA, and the elevation of lean mass and food intake. After a few years, Wong et al (2015) compared the potential of αTF, αT3, γT3, and δT3 at the dosage of 85 mg/kg/day in improving metabolic conditions. The findings showed the superiority of δT3 among the treatments in minimizing liver injuries, reducing total fat mass, abdominal circumference, adiposity index, retroperitoneal fat, epididymal fat, FBG, systolic BP, TC, NEFA, and TG levels.…”

“…Therefore, dietary supplementation of tocotrienol preparations with minimal concentrations of tocopherol (<20%) in the mixture has been recommended for the inhibition of several biological activities [12,20]. All the isomers of tocols were detected in plasma as well as in various tissues of rats (liver, heart, and adipose) [21] and humans (skin, adipose, brain, cardiac muscle, and liver) [22]. Moreover, high doses of tocotrienols may be useful for cancer chemoprevention and treatment.…”
