Anti‐influenza virus agents: Synthesis and mode of action

Abstract: Annual epidemics of influenza virus infection are responsible for considerable morbidity and mortality, and pandemics are much more devastating. Considerable knowledge of viral infectivity and replication has been acquired, but many details still have to be elucidated and the virus remains a challenging target for drug design and development. This review provides an overview of the antiviral drugs targeting the influenza viral replicative cycle. Included are a brief description of their chemical syntheses and … Show more

“…However, numerous medicinal chemical efforts directed toward discovery of small-molecule inhibitors of resistant M2 mutants (18)(19)(20)(21) have shown little progress until recently (4,22,23). For example, we designed a spiroadamantane inhibitor of V27A and L26F mutants (23), but this compound did not inhibit S31N.…”

“…This approach, as well as others, has led to the development of highly selective therapies for the treatment of viral infections and has been reviewed previously (5,9,20). While these therapeutic agents have excellent activities against the target enzymes from specific viruses, they can have reduced or poor efficacy against other closely related viruses.…”

“…piperidine, 2-(1-adamantyl-2-methyl)pyrrolidine, and many others [77][78][79][80][81][82][83][84][85] (as reviewed by Lagoja and De Clercq 86 ). 87 In all cases, the anti-influenza A virus activity noted for these compounds compared favorably to that of amantadine when evaluated under the same experimental conditions.…”
