Curcumin, a dietary pigment in curry, suppresses tumor initiation and tumor promotion. Curcumin is also a potent inhibitor for AP-1 and NF-kB activation. In this report, we show that curcumin inhibits JNK activation by various agonists including PMA plus ionomycin, anisomycin, UV-C, g radiation, TNF-a, and sodium orthovanadate. Although both JNK and ERK activation by phorbol 12-myristate 13-acetate (PMA) plus ionomycin were suppressed by curcumin, the JNK pathway was more sensitive. The IC 50 (50% inhibition concentration) of curcumin was between 5 ± 10 mM for JNK activation and was 20 mM for ERK activation. In transfection assays, curcumin moderately suppressed MEKK1-induced JNK activation; however, it e ectively blocked JNK activation caused by co-transfection of TAK1, GCK, or HPK1. Curcumin did not directly inhibit JNK, SEK1, MEKK1 or HPK1 activity. Although curcumin suppressed TAK1 and GCK activities at high concentrations, this inhibition cannot fully account for the JNK inhibition by curcumin in vivo. Our data suggest that curcumin may a ect the JNK pathway by interfering with the signaling molecule(s) at the same level or proximally upstream of the MAPKKK level. Taken together, the inhibition of the MEKK1-JNK pathway reveals a possible mechanism of suppression of AP-1 and NF-kB signaling by curcumin, and may explain the potent anti-in¯ammatory and anti-carcinogenic e ects of this chemical.

Excited-State Intramolecular Hydrogen Atom Transfer and Solvation Dynamics of the Medicinal Pigment Curcumin

The potential use of the naturally occurring yellow-orange pigment curcumin as a photodynamic therapy agent is one of the most exciting applications of this medicinal compound. Although subnanosecond spectroscopy has been used to investigate the photophysical processes of curcumin, the time resolution is insufficient to detect important and faster photoinduced processes, including solvation and excited-state intramolecular hydrogen atom transfer (ESIHT). In this study, the excited-state photophysics of curcumin is studied by means of ultrafast fluorescence upconversion spectroscopy. The results show two decay components in the excited-state kinetics with time scales of 12−20 ps and ∼100 ps in methanol and ethylene glycol. The resulting prominent isotope effect in the long component upon deuteration indicates that curcumin undergoes ESIHT in these solvents. The short component (12−20 ps) is insensitive to deuteration, and multiwavelength fluorescence upconversion results show that this decay component is due to solvation of excited-state curcumin. A 1064 nm Dispersive Raman Spectral Imaging System for Food Safety and Quality Evaluation

Raman spectral imaging is an effective method to analyze and evaluate the chemical composition and structure of a sample, and has many applications for food safety and quality research. This study developed a 1064 nm dispersive Raman spectral imaging system for surface and subsurface analysis of food samples. A 1064 nm laser module is used for sample excitation. A bifurcated optical fiber coupled with Raman probe is used to focus excitation laser on the sample and carry scattering signal to the spectrograph. A high throughput volume phase grating disperses the incoming Raman signal. A 512 pixels Indium-Gallium-Arsenide (InGaAs) detector receives the dispersed light signal. A motorized positioning table moves the sample in two-axis directions, accumulating hyperspectral image of the sample by the point-scan method. An interface software was developed in-house for parameterization, data acquisition, and data transfer. The system was spectrally calibrated using naphthalene and polystyrene. It has the Raman shift range of 142 to 1820 cm −1 , the spectral resolution of 12 cm −1 at full width half maximum (FWHM). The spatial resolution of the system was evaluated using a standard resolution glass test chart. It has the spatial resolution of 0.1 mm. The application of the system was demonstrated by surface and subsurface detection of metanil yellow contamination in turmeric powder. Results indicate that the 1064 nm dispersive Raman spectral imaging system is a useful tool for food safety and quality evaluation.
