Mangrove forest is a natural ecosystem that has been designated as a conservation and tourism area. Besides its function as an ecotourism area, mangrove forests also store biological wealth in the form of bacteria associated with soil from mangrove plants. Bacteria from mangrove communities are known to synthesize various secondary metabolites. However, the bacterial biodiversity associated with soil from mangrove plants, especially those in Bali, has not been explored optimally. This study was designed to analyze the potential of antioxidant activity and cytotoxicity of five bacterial isolates namely Bacillus sp. SA1, Bacillus sp. SA4, Bacillus sp. AM23, Bacillus sp. RM10 and Bacillus sp.RM 18 which were previously isolated from mangrove soil in Ngurah Rai mangrove forest Denpasar Bali. These five bacterial isolates were grown on ISP-2 liquid media and were fermented for seven days. Subsequently, the supernantant of each bacterial isolates was extracted using 100 mL of ethyl acetate with 1:1 ratio (v:v). The antioxidant activity of each ethyl acetate extract was analyzed in vitro using DPPH (1,1-diphenyl-2-picrylhydrazil) assay and the cytotoxicity activity was evaluated by the Brine Shrimp Lethality assay. Results showed that ethyl acetate extracts of the five bacterial isolates display weak antioxidant activity with IC50 values > 150 ppm. The cytotoxicity test showed that the etyhyl acetate crude extract of bacterial isolate RM10 inhibted 70% of Artemia salina larvae, followed by crude extract of Bacillus sp. RM18 and Bacillus sp. SA4 isolate extracts which equally inhibited 60% of these larvae. This results indicated the potential of isolate Bacillus sp.RM18, Bacillus sp.RM10 and Bacillus sp. SA4 in producing cytotoxic compounds. GC-MS chemical profiling of ethly acetate Bacillus sp. RM10 showed 11 different compounds. Three compounds namely 5hydroxymethylfurfural, 1,3,-propanediamine and 2-Aminononadecane were the most dominant compounds detected in the extract. Overall, these findings confirmed the potential bacteria associated with mangrove soil as the source of bioactive compounds.

Seaweeds like other marine organisms develop a strong association with prokaryotic communities, especially bacteria. Many of these associated bacteria play an important role in synthesizing secondary metabolites which are beneficial for their host, especially against predation or infection. In Indonesia, Eucheuma cottonii is one of the seaweed species that has been cultivated widely. However, only limited study has been reported on bacterial isolates and their antibacterial activity from E. cottonii. Bacterial isolates from E. cottonii were cultivated in six agar media namely Zobell Marine Agar, Nutrient Agar, Plate Count Agar, ISP-1, ISP-2, and Starch-M protein agar. Cultivation of bacterial isolates yielded 23 isolates with nine bacterial isolates were classified as Gram-positive bacteria and 14 isolates were grouped among Gram-negative bacteria. All the bacterial isolates were screened for their antibacterial activity against six bacterial indicator strains namely Staphylococcus aureus ATCC 25923, Streptococcus mutans FNCC 0405, Escherichia coli ATCC 25922, and Klebsiella pneumoniae ATCC 700603 using perpendicular streak and agar block method. Six out of 23 bacterial isolates displayed antibacterial activity against at least one of the bacterial indicator strains. The bacterial isolate ISP1RL4 showed the highest antibacterial activity with average inhibition of > 20 mm against all bacterial indicators. Overall, our result indicated the potential of bacterial isolates associated with E. cottonii as an antibacterial producer.

Resistensi bakteri merupakan isu global yang sangat penting. Infeksi penyakit disebabkan oleh resistensi bakteri. E. coli, S. aureus dan S. thypi merupakan bakteri patogen yang menyebabkan infeksi. Pengendalian resistensi bakteri dapat dilakukan dengan eksplorasi sumber senyawa baru. Bakteri sedimen mangrove berpotensi sebagai antibakteri terhadap E. coli, S. aureus dan S. thypi. Tujuan dari penelitian ini dilakukan untuk memperoleh jenis bakteri sedimen mangrove yang memiliki aktivitas antibakteri terhadap E. coli, S. aureus dan S. thypi. Penelitian terdiri dari beberapa tahapan dimulai dari isolasi, purifikasi, uji aktivitas antibakteri dan uji biokimia. Hasil isolasi yang diperoleh berjumlah 33 isolat, 3 diantaranya memiliki aktivitas antibakteri terhadap E. coli, S. aureus dan S. thypi. Berdasarkan hasil yang didapatkan, Aktivitas antibakteri SS.2 ISP tergolong kuat terhadap E. coli dan tergolong sedang terhadap S. aureus dan S. thypi. Aktivitas antibakteri SS.4 ISP tergolong sedang terhadap E. coli, S. aureus dan S. thypi sedangkan SK.1 MA tergolong sedang terhadap E. coli dan S. aureus dan tergolong lemah terhadap S. thypi. Berdasarkan identifikasi biokimia isolat SS.2 ISP, SS.4 ISP dan SK.1 MA teridentifikasi sebagai genus Pseudomonas sp., Moraxella sp. dan Vibrio sp.. Bacterial resistance is a very important global issue. Infectious diseases are caused by bacterial resistance. E. coli, S. aureus and S. thypi are pathogenic bacteria that cause infection. Control of bacterial resistance can be done by exploring new sources of compounds. Mangrove sediment bacteria have the potential as antibacterial against E. coli, S. aureus and S. thypi. The purpose of this study was to obtain types of mangrove sediment bacteria that have antibacterial activity against E. coli, S. aureus and S. thypi. The research consisted of several stages starting from isolation, purification, antibacterial activity test and biochemical test. The isolation results obtained were 33 isolates, 3 of which had antibacterial activity against E. coli, S. aureus and S. thypi. Based on the results obtained, the antibacterial activity of SS.2 ISP was strong against E. coli and moderate against S. aureus and S. thypi. The antibacterial activity of SS.4 ISP was moderate against E. coli, S. aureus and S. thypi, while SK.1 MA was moderate against E. coli and S. aureus and weak against S. thypi. Based on the biochemical identification of isolates SS.2 ISP, SS.4 ISP and SK.1 MA were identified as the genus Pseudomonas sp., Moraxella sp. and Vibrio sp.
