Background and Aim: The need for fish seeds resistant to bacterial and viral infections has encouraged studies on the molecular pathogenesis mechanism of Vibrio bacteria, such as Vibrio alginolyticus and Vibrio harveyi, regarding the receptor organs, protein adhesion mechanisms, and antibody responses of the humpback grouper. This study aims to confirm the characteristics of the specific proteins expressed in the receptor organ of the humpback grouper (Cromileptes altivelis) using the expression of V. alginolyticus and V. harveyi bacteria. Materials and Methods: The study was conducted by isolating crude protein and whole cells from both the Vibrio bacteria. In addition, serum and organ tissue were also isolated from fish samples. Then, hemagglutination and dot blot tests with polyacrylamide gel electrophoresis analysis were performed to determine the highest expression of receptor from the whole bacterial cells and crude protein from both healthy and infected (V. alginolyticus and V. harveyi) fishes. Scanning electron microscope results showed that V. alginolyticus and V. harveyi could express bundle-forming pili, which is involved in bacterial autoaggregation and the mediation of the initial attachment of bacteria to their host cells. Results: These results indicated that all the specific receptors for protein in fish organs recognized vibriosis antigens. The specificity test showed that the brain, eye, and kidney organs' receptors provided a quality and quantity level of responses at 22.63, 53.95, and 43.15 kDa, respectively. The polyclonal anti-V. alginolyticus immunoglobulin M (IgM) antibodies were more cross-reactive than the anti-V. harveyi IgM. Hence, this shows that V. alginolyticus bacteria are more pathogenic than V. harveyi. Conclusion: In the future, the molecular characteristics of V. alginolyticus and V. harveyi antigens and the specific receptor organ proteins in the humpback grouper can be developed as the basis for constructing molecular peptide-based vaccine materials.

Role of Pili Protein 11 kDa ofÂ Streptococcus pneumoniaeÂ as HemagglutininÂ and Adhesin ProteinÂ Streptococcus pneumoniae has pili which play roles in adhesion, colonization of nasopharyngeal epithelial cells, and phagocytic inhibition of immune cells. This study aimed to determine the characteristics of the 11 kDa pili protein as hemagglutinin and adhesin, as well as their immune responses. The 11 kDa pili protein from S. pneumoniae was isolated by SDS-PAGE, purified by electroelution and dialysis. Hemagglutination and adhesion tests were carried out on the protein, and western blotting of the polyclonal antibody immune responses were evaluated. Hemagglutination test showed that the 11 kDa pili protein played a role in the hemagglutination process up to 2-time dilution. Adhesion test showed there was a correlation between the dose of the protein and the bacteria attached to the epithelial cells. The Pearson correlation test showed a P value of 0.010 and a correlation coefficient of R = -90.919. Quadratic regression test produced R2 = 0.974. Western blotting test showed that 11 kDa pili protein polyclonal antibodies recognized 67 kDa and 11 kDa pili proteins. The study concluded that the 11 kDa S. pneumoniae pili protein acted as hemagglutinin and adhesin, and the polyclonal antibody protein responded to 67 pDa and 11 kDa BM pili proteins.Keywords: adhesin, hemagglutinin, pili, protein 11 kDa, Streptococcus pneumoniaeÂ ABSTRAKStreptococcus pneumoniae memiliki pili yang berperan dalam adhesi, kolonisasi sel epitel nasofaring, serta sebagai inhibitor fagositosis sel imun. Introduction: Streptococcus pneumoniae (S. pneumoniae) causes pneumococcal disease, which has high mortality and morbidity in children under two years of age, the elderly and immunocompromised individuals. This disease can be prevented by immunization, but the current vaccine, pili protein vaccine (PPV), is less likely to protect children under the age of two and only protects against the serotypes contained in the vaccine. Hence, a new vaccine is needed to enable full protection. The use of bacterial pili proteins may offer an alternative new vaccine. Therefore, the determination of the ability of such proteins to stimulate mucosal immunity with indicator expression of pIgR and s-IgA is required. Materials and Methods: Pili were isolated using the pili bacterial cutter method, and used for nasal vaccination to the rats. TGF-β1, IL-17A, and s-IgA were measured by ELISA while pIgR was examined by immunohistochemistry. Results: This study demonstrated that intranasal immunization with antigen (54 kDa pili protein) and antigen plus adjuvant significantly increased (p<0.05) the expression of TGF-β1. However, the expression of IL-17A increased significantly (p<0.05) only in rats immunized with antigen plus adjuvant. Further analysis demonstrated that intranasal immunization with antigen and antigen plus adjuvant significantly increased (p<0.05) expression of pIgR. Expression of sIgA in nasal lavage significantly increased (p<0.05) in those rats which had been immunized with pili protein plus adjuvant. Conclusion: This study showed that the immünization with S. pneumoniae pili protein increased expression of pIgR and sIgA which are important mucosal immunity components. Therefore, this protein has potency to be developed as nasal vaccination to prevent S. pneumoniae infection.
