Anticholinergic and antioxidant activities of avocado (Folium perseae) leaves – phytochemical content by LC-MS/MS analysis

Abstract: In the first stage of the manuscript, we aimed to examine antioxidant capacity and anticholinergic properties of avocado (Folium perseae) leaves. Avocado leaf was extracted by water (WEFP) and ethyl alcohol (EEFP) and antioxidant activity was determined using by several antioxidant assays including DPPH· and ABTS •+ radical scavenging assays, Cu 2+ -Cu + reducing, Fe 3+ -Fe 2+ reducing, and FRAP reducing activities. Avocado leaf extracts demonstrated antioxidant activity and anticholinergic activities, while α… Show more

“…The FRAP method is a simple, cheap, and fast method that is used to determine the antioxidant capacity of many phenolic, flavonoid, and similar molecules under in vitro conditions. [ 37,56–58 ] Using this method, we analyzed the reduction of Fe 3+ by naphthol‐derived molecules. Fe 2+ in a reduced form gives absorbance at 593 nm by forming a colored complex with TPTZ.…”

“…As with DPPH• radical scavenging activity, the ABTS •+ radical scavenging activity method is widely used in pure substances, extracts, and beverages. [ 37,57,58 ] In this method, ABTS •+ cation radical must be created first. The method is based on the direct formation of the blue‐green colored ABTS •+ solution from the interaction of ABTS and K 2 S 2 O 8 and the measurement of this cation radical spectrophotometrically.…”

“…Cu 2+ -reducing ability of EEFO and WEFO and standards at the same concentration (30 μg/mL) demonstrated the following order: BHT (λ700: 1.561±0.089, r 2 : 0.9978) >ascorbic acid (λ700: 1.069±0.007, r 2 : 0.9722) > α-tocopherol (λ700: 0.785±0.061, r 2 : 0.9986) > EEFO (λ700: 0.954±0.017, r 2 : 0.9928) and WEFO (λ700: 0.236± 0.020, r 2 : 0.9998). The iron-chelating ability is very significant due to ionic species like ferrous ion (Fe 2+ ) facilitating the production of free ROS in the organism [67]. This binding assay is a significant antioxidant method used for prevention or delaying of oxidation processes catalyzed by metal ions, however excessive metal ions can cause cell damage.…”

“…With all this, it is well established that polyphenols also have anti-AD properties and α-glycosidase inhibition profiles. Therefore, one of the most important therapy approaches for neurodegenerative diseases and DM is natural compounds and products including phenolic compounds [28,29,30,31]. However, current evidence demonstrates that patients with DM have an increased risk of developing AD.…”
