Botrytis cinerea is the causal agent of grey mould rot of apples. The efficacy of biofumigation with thyme (Thymus vulgaris), savoury (Satureja montana), and basil (Ocimum basilicum) essential oils (EOs) at 1%, 0.5%, and 0.1% concentrations were tested against B. cinerea. In vitro, the results showed 100% growth inhibition at 1% concentration for all oils. Subsequent biofumigation experiments on apples of cultivar ‘Opal’ with 1% EOs showed that, after 60 d storage, thyme and savoury EOs significantly reduced grey mould rot incidence (average incidence 2% for both treatments) compared to the control (7%). Analyses of quality indicated slightly higher fruit firmness for 1% thyme at 30 d and slightly higher titratable acidity for 1% thyme and savoury at 60 d. Sampling of the atmosphere inside the cabinets was performed to characterize and quantify the volatile components of EOs released through biofumigation. Though thymol and p-cymene were the main components of thyme EO, the antimicrobial activity was mainly due to the presence of thymol and, to a lower extent, of carvacrol. In savoury EO, carvacrol and p-cymene were the main components, whereas in basil EO, linalool and estragole were mainly present. Metabarcoding analyses showed that the epiphytic microbiome had higher richness and evenness compared to their endophytic counterpart. By the end of shelf-life, treatments with thyme EO reduced B. cinerea abundance compared to the inoculated control for both endophytes (from 36.5% to 1.5%) and epiphytes (from 7.0% to 0.7%), while favouring a significant increase in Penicillium species both in endophytes (from 0.2% to 21.5%) and epiphytes (from 0.5% to 18.6%). Results indicate that thyme EO (1%) and savoury EO (1%) are equally effective in hampering grey mould rot development in vivo.

Introduction. Fungal genera Alternaria and Fusarium include human and plant pathogenic species. Several antifungals have been used for their control, but excessive use has contributed to resistance development in pathogens. An alternative to searching for and developing new antifungal agents is using essential oils and their main components, which have biological activities of interest in medicine and food production.Objective. To evaluate in vitro and in silico the antifungal activities of terpenoids against Alternaria alternata and Fusarium oxysporum.Materials and methods. The minimum inhibitory concentration and minimum fungicidal concentration values of 27 constituents of essential oils used against Alternaria alternata and Fusarium oxysporum were evaluated in vitro. In addition, using genetic algorithms, quantitative models of the structure-activity relationship were used to identify the structural and physicochemical properties related to antifungal activity.Results. The evaluated compounds proved to be effective antifungals. Thymol was the most active with a minimum inhibitory concentration of 91.6 ± 28.8 μg/ml for A. alternata and F. oxysporum. Quantitative structure-activity relationship models revealed the octanolwater cleavage ratio as the molecular property, and the phenols as the main functional group contributing to antifungal activity.Conclusion. Terpenoids exhibit relevant antifungal activities that should be incorporated into the study of medicinal chemistry. Inclusion of in silico assays in the in vitro evaluation is a valuable tool in the search for and rational design of terpene derivatives as new potential antifungal agents.

Bu çalışma “Golden Delicious” elma çeşidinde Kurşuni küfe (Botrytis cinerea Pers.) karşı koruyucu ve iyileştirici etkide uçucu yağların (UY) bireysel ve kombinasyonlarının besin elementlerinin içeriği üzerine ilişkileri değerlendirmek amacı ile yürütülmüştür. Bu amaca yönelik çalışmada enfeksiyon öncesi (koruyucu etki) ve sonrası (iyileştirici etki) elma meyveleri kullanılarak timol, ögenol, 1,8-sineol UY’ların koruyucu ve iyileştirici özelliğinin besin elementleri içeriğine etkileri araştırılmıştır. Hasat edilen elmalar UY (tekli kombinasyonlarda 1,25 μL, ikili kombinasyonlarda 2,5 μL, üçlü kombinasyonlarda 3,75 μL) içeren çözeltilere batırılarak 30 dakika boyunca inkube edilip ±4°C'de 7 gün depolanmıştır. Daha sonra Kjeldahl yöntemi ve bir Vapodest Hızlı Kjeldahl Distilasyon Ünitesi kullanılarak makro ve mikro besin elementleri belirlenmiştir. Sonuçlara göre, B. cinerea ve B. cinerea’ya karşı UY uygulamalarının besin elementi içeriği üzerine olumlu etki yaparak meyve kalitesini koruduğu tespit edilmiştir. Uygulama yöntemlerinden iyileştirici etkinin koruyucu etkiye göre besin elementleri içeriğinin kontrol grubuna göre arttırmada daha etkili olduğu gözlemlenmiştir. Ayrıca UY’ların bireysel ve kombinasyonalarının besin elementleri içeriğine etkileri incelendiğinden en fazla timol ve ögenol kombinasyonlarının oluşturduğu gruplarda içeriği en fazla arttırdığı belirlenmiştir. Tüm besin elementleri içeriği değerlendirildiğinde ise, Na içeriğinin en yüksek olduğu buna karşın kükürttün diğer elementlerden daha düşük içeriğe sahip olduğu saptanmıştır. Sonuç olarak hasat sonrası meyvelerde B. cinerea’ya karşı UY uygulamasının makro ve mikro besin elementleri yönünden iyi sonuçlar elde edilmiştir. Elmalarda hasat sonrası kurşuni küfe karşı uçucu yağların sentetik fungusitlerin yerine alternatif olacağını ve gelecek açısından ümit verici olduğunu düşünmekteyiz
