Antigenic determinants of heavy chain variable regions: immunological typing of the human immunoglobulin VHIII subgroup

Abstract: An antigenic determinant of the VHIII variable region subgroup was defined by means of a heterologous specific antiserum using a hemagglutination inhibition procedure. The specificity of this antiserum was established in inhibition experiments with proteins either of known primary structure or belonging to a definite VH subgroup. A series of IgG, IgA, IgM and IgD monoclonal proteins was examined for the presence of this VHIII subgroup antigenic determinant. The data showed that 50% of the IgG, 62% of the IgA, … Show more

“…The heavy chains of proteins Shm, Slo, Dov, and Yos had pyrrolidonecarboxylic acid (PCA)-blocked NHz-termini. Serological typing using a VHIII/anti-VnIII hemagglutination inhibition system [25] indicated that the Shm heavy chain was VHIII, whereas the other three were non-VHIII.…”

“…Fur thermore, anti-D sensitized RBC were tested with the following antiglobulin sera: antiIgG4, anti-IgA, anti-IgAl, anti-IgA2 and anti-IgM. Antisera preparation and typing methods were previously described [9], All these tests were negative.…”
