Antihypertensive Action of Allantoin in Animals

Abstract: The agonists of imidazoline I-1 receptors (I-1R) are widely used to lower blood pressure. It has been indicated that guanidinium derivatives show an ability to activate imidazoline receptors. Also, allantoin has a chemical stricture similar to guanidinium derivatives. Thus, it is of special interest to characterize the effect of allantoin on I-1R. In conscious male spontaneous hypertensive rats (SHRs), mean blood pressure (MBP) was recorded using the tail-cuff method. Furthermore, the hemodynamic analyses in c… Show more

“…Allantoin has been known to enhance the efficacy and desirability of cosmetic creams with antioxidative and antiinflammatory activities. Recent researches also indicate that allantoin has the effects of antidiabetic, memory‐enhancing, and antihypertension (Niu and others ; Chen and others ; Go and others ). To effectively exploit the value of yam and yam peel on health protection, it is significant to investigate the principal component in yam and yam peel, which have the effect of antioxidant activity in vitro and antitumor in vivo .…”

“…However, there has been no trial conducted to investigate the antitumor activity, especially the inhibition effect of CYP which contains more bioactive substance such as allantoin and flavonoids. Allantoin (hydantoin 5‐ureido) has been widely described in literatures as for numerous pharmacological activities, including antiinflammatory, anti‐irritating, antidiabetic, and antihypertension (Shestopalov and others ; Chen and others ; Go and others ). In recent years, increased investigations have focused on the bioactivities of hydantoins, such as antiproliferative, cytotoxicity, and anticancer (Puszyńska‐Tuszkanow and others ).…”

“…Allantoin can activate I-1R in brain to produce antihypertension [47] and decrease feeding behaviors [48]. Allantoin can stimulate I-2R to lower blood sugar and improve insulin sensitivity in type 1-like diabetic rats [39][40][41][42].…”

“…In this study, it was hypothesized that, because of its ability to cross the blood–nerve barrier and reach the areas of dorsal root ganglions and nerve terminals affected by cisplatin, allantoin has an anti‐oxidant activity, reducing the potential toxic effect of cisplatin and thus reducing degeneration of the small nerve fibers. In a literature survey, allantoin has been reported to possess anti‐diabetic, anti‐hypertensive, and analgesic activity…”
