Antimalarial Drug Artemisinin Extenuates Amyloidogenesis and Neuroinflammation in APPswe/PS1dE9 Transgenic Mice via Inhibition of Nuclear Factor‐κB and NLRP3 Inflammasome Activation

Abstract: The in vivo study clearly demonstrates that artemisinin has protective effects on AD pathology due to its effects on suppressing NF-κB activity and NALP3 inflammasome activation. Our study suggests that targeting NF-κB activity and NALP3 inflammasome activation offers a valuable intervention for AD.

“…As we know, neuroinflammation played a major role in the formation of Aβ; besides, activated Nrf2 activity also shows benefits to AD in reports [42]. In addition, the reports have showed that artemisinin decreased BACE1 expression, and suppressed NF-kB activity and NALP3 activation [43]. Furthermore, it just reported that artemisinin was able to modulate GABA (A) receptor signaling, while the GABA(A) receptor signaling was seriously altered in aging and AD [44], [45].…”

“…In addition, studies have showed that NFκB/MAPK signaling contributes to inflammasome assembly activation, i.e. by maturation of IL‐1 and IL‐18 following cleaved procaspase‐1 by oligomerzied NLRs, adaptor protein ASC, and procaspase‐1, eventually leading to downstream cascade of inflammatory reaction . Cytokine IL‐6 plays a direct role in inflammation and neurodegenerative diseases.…”

“…There has been no report about the therapeutic effect of artesunate on neurodegenerative disorders. However, study clearly demonstrated that artemisinin had protective effects on Alzheimer's disease pathology through suppressing NF- κ B activity and NALP3 inflammasome activation in APPswe/PS1dE9 transgenic mice [93]. As the water-soluble derivatives of artemisinin, the protective effects of artesunate on Alzheimer's disease or even neurodegenerative disorders merit in-depth studies.…”
