Antimicrobial and Enzyme Activity Produced by Bacillus Spp. Isolated From Soil

Abstract: Objective: Our study was to isolate Bacillus spp. from soil all around 6 geographic parts of Thailand and screen for potential antimicrobial. Methods: A total of 43 isolates which isolated from 100 samples of soil were investigated. Preliminary screening was based on antimicrobial activity against 16 strains of pathogenic bacteria and fungi, including 10 strains of Methicillin-resistant Staphylococcus aureus (MRSA), and 1 strain of Methicillin-sensitivity Staphylococcus aureus (MSSA), Extended spectrum beta lac… Show more

“…After culturing on the medium and incubation at the optimal temperature, the plate was floated by 0.2% Congo red and thus decolorized by NaCl 1 mol l −1 for 15 min. The existence of cellulase activity was indicated by a prominent halo surrounding the developing strain ( 22 , 23 ).…”

“…The bacterial isolates were tested, using the spot agar method, for the following enzymatic activities: amylolytic activity (starch hydrolysis), esterase activity, proteolytic activity (protease and gelatinase), and hemolytic activity. All assays were carried out using basal mineral salts medium (BMSM) with a specific substrate and in triplicate [ 28 ].…”

“…Bacterial isolates were spotted and incubated at 37 °C for 2 days. The presence of clear zones around the colonies indicated positive esterase activity [ 28 ].…”

“…Antimicrobial activity was tested by agar well diffusion method. Under Joint FAO/WHO Expert Committee on Food (JECFA) procedures [ 47 ] and the study carried out by Powthong & Suntornthiticharoen [ 48 ], nine different bacteria were used as indicators to verify the antimicrobial capacity of the Bacillus spp. isolated from the gut microbiota.…”
