The aim of this study was to examine the antioxidant activities of Physalis peruviana L. (Solanaceae) aqueous extract (PPWE) and its protective effect against acetaminophen (APAP)-induced hepatotoxicity in rats. Using different models of antioxidant assay, namely ferric thiocyanate, 2,2-diphenyl-1-picrylhydrazyl (DPPH), and reducing power, PPWE showed a dose-dependent increase in antioxidant activities, with total antioxidant activity (IC 50 : 0.81 µg/ml) close to that of vitamin C (IC 50 : 0.89 µg/ml). APAP at 850 mg/kg significantly increased the levels of serum glutamic pyruvic transaminase (sGPT), glutamic oxaloacetic transaminase (sGOT) and alkaline phosphatase (sALP). However, pre-treatment with PPWE at doses150, 300, and 600 mg/kg body weight significantly prevented the increase in these enzymes, which are the major indicators of liver hepatitis. Biochemical assays of liver homogenate showed that PPWE at 150∼600 mg/kg significantly enhanced superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx) concentrations, and diminished the level of thiobarbituric acid reactive substances (TBARS). Furthermore, liver histological observation also showed an obvious amelioration in the liver cell necrosis, liver lesion, and fatty changes in PP-treated groups. High performance liquid chromatographic analysis showed that ellagic acid (ca. 0.2%) but not others could be the major component contributing to the antioxidant and hepatoprotective activities of PPWE. The present study concludes that PPWE possesses antioxidant activity and potent hepatoprotective effect against APAP-induced liver injury in rats.

ÖzetAmaç: Antioksidan aktivite gösteren ve eser elementlerin gerekli miktarları-nı içeren tıbbi gıdaların diyetle alınması, iyi bir sağlıklı yaşam sürdürmek için önemlidir. Çalışmamızda, Kayseri'de marketlerde satılan Altınçilek (Physalis peruviana L.) meyvesinin antioksidan kapasitesinin ve metal içeriğinin belirlenmesi amaçlanmıştır. Gereç ve Yöntem: P. peruviana meyvesinin antioksidan kapasitesi, 2,2-difenil-1-pikrilhidrazil (DPPH) serbest radikal süpürme testi ile belirlenmiştir. Meyvenin metanol (MeOH) ekstresinin DPPH serbest radikalini temizleme kapasitesi, referans olarak askorbik asit, gallik asit ve bütillen-dirilmiş hidroksitoluen (BHT) gibi bilinen antioksidanlarla karşılaştırılmıştır. P. peruviana meyvesinin metal içeriği atomik absorpsiyon spektrometresi (AAS) kullanılarak ölçülmüştür. Bulgular: P. peruviana meyve ekstresinin DPPH serbest radikal temizleme aktivitesinin olduğu, fakat antioksidan kapasitesinin standart maddelerden daha düşük olduğu tespit edilmiştir. P. peruviana, askorbik asit, gallik asit ve BHT'nin % 50 inhibitör konsantrasyon (IC 50 ) değer-leri sırası ile 32 mg/ml, 3.8 mg/ml, 3.51 mg/ml ve 1.21 mg/ml olarak belirlenmiştir. AAS ile yapılan analizler sonucu, meyvenin eser element yönünden zengin içeriğe sahip iken ağır metal içeriğinin az miktarda veya hiç olmadı-ğı gözlenmiştir. Tartışma: Bu sonuçlar, P. peruviana meyvesinin doğal kaynaklı potansiyel bir antioksidan ve eser element kaynağı olduğunu göstermektedir. The DPPH radical scavenger capacity of the methanol extract of fruit was compared with known antioxidants such as ascorbic acid, gallic acid, and butylated hydroxytoluene (BHT) as references. The metal content of P. peruviana fruit was measured by using atomic absorption spectrometer (AAS). Results: The fruit of P. peruviana was found to possess DPPH free radical scavenging activity but the antioxidant capacity was lower than the standard substances. Inhibitory concentration 50% (IC 50 ) values of P. peruviana, ascorbic acid, gallic acid, and BHT were determined as 32 mg/ml, 3.8 mg/ml, 3.51 mg/ ml, and 1.21 mg/ml, respectively. As a result of the analysis by AAS, it was observed that P. peruviana fruit contented plentiful trace elements and the content of heavy metal was small amount or not detected. Discussion: These observations suggest that the fruit of P. peruviana has a potential source of antioxidant and trace elements of natural origin.

ResumoO trabalho foi conduzido em casa de vegetação e objetivou analisar o crescimento e a partição de assimilados em Physalis peruviana submetida a intervalos de adubação foliar. As plantas foram coletadas a intervalos regulares de quatorze dias após o transplante até o final do ciclo e foram determinados a massa seca e a área foliar. A partir dos dados primários foi aplicada a análise de crescimento, sendo calculados a massa seca total (W t ), taxas de produção de matéria seca (C t ), crescimento relativo (R w ) e assimilatória liquída (E a ), índice de área foliar (L), razões de área foliar (F a ) e massa foliar (F w ), área foliar específica (S a ), partição de matéria seca entre órgãos e o número de frutos. Plantas de Physalis peruviana submetidas à adubação foliar quinzenal atingiram maior W t , C t , superior número de frutos e semelhante matéria seca de frutos (W fr ) a plantas submetidas à aplicação de adubação foliar semanal e superior W fr em relação a plantas sem adubação foliar. Desse modo, a aplicação de adubação foliar proporcionou benefícios no crescimento e na partição de assimilados em plantas de Physalis peruviana. Palavras-chave: Physalis peruviana, área foliar, matéria seca, taxas de crescimento AbstractThe work was conducted in greenhouse and aimed to analyze the growth and partitioning of assimilates in Physalis peruviana subjected in intervals of leaf fertilization. The plants were collected at regular intervals of fourteen days after transplantation until the end of the cycle and determined the dry mass and leaf area. From the primary data analysis was applied to growth analysis and calculated the total dry matter production (W t ), rates of dry matter production (C t ), relative growth (R w ), net assimilation (E a ), leaf area index (L), relative growth of leaf area (F a ) and ratios of leaf area and leaf mass (F w ), specific leaf area (S a ) and dry matter partitioning between organs. Plants of Physalis peruviana subjected to leaf fertilization biweekly reached higher W t , C t , number of fruits and similar dry mass of fruits (W fr ) of plants subjected to foliar weekly application and higher W fr compared to plants without application leaf of fertilization. Thus, the application of leaf fertilization provided benefits of growth and the partition of assimilates in Physalis peruviana plants.
