Antioxidant- Influenced Alterations in Glutathione Reductase Activity in Different Age Groups of Male Mice

Abstract: The glutathione reductase activity was estimated in brain and liver of male mice of different age groups fed on butylated hydroxy anisole (BHA), propyl gallate (PG) and α-tocopherol (TP). A significant increase in glutathione reductase activity was observed with TP during all the phases (pre-reproductive, reproductive and post-reproductive). Antioxidants enhanced the enzyme activity in both the tissues with the exception of BHA in brain during reproductive and PG in liver during pre-reproductive phases respect… Show more

“…The increase in enzyme activty during reproductive phase could be due to increased metabolic rate during this phase of life resulting in increased production of free radicals and peroxides which need detoxification. These results corroborate with Khanna et al [36] who found an increase in GR activity during reproductive phase in male mice. A gradual decline with age was observed in GR activity of both the sexes.…”

“…The flies were homogenized in suitable buffer (as described in each assay) using Polytron homogenizer at 4"C for 30 sec (2x15 sec with 15 sec cooling interval). Estimations of antioxidative enzymes (catalase, peroxidase and glutathione reductase) were done on whole body homogenates of males and females at 1,8,15,22,29,36 and 43 days of age.…”

“…The induction of GR has been demonstrated with other antioxidants such as butylated hydroxyanisole, propyl gallate, ·-tocopherol [6], lobenzarit (disodium 4-chloro-2,2-iminodibenzoate) [24], and fraxetin (6-methoxy-7,8-dihydroxycoumarin) [25].…”

“…Based on this theory, experimental gerontologists, particularly those attempting to retard senescence by environmental manipulation, include the use of nutrients or drugs that increase antioxidants in gerontological research [5]. The enhanced activity of glutathione reductase (GR) by treatment with phenolic antioxidant compounds, such as 3,5-di-terc-butyl-4-hydroxytoluene (BHT), appears to be indicative of an enhanced cellular reducing potential for protecting the organism against oxidative damage [6]. Moreover, studies have shown that mice treated with this synthetic phenolic antioxidant under optimal living conditions have a significant increase in median life span [7].…”

“…The decrease in the GSSG concentration observed in PDL 50→75 cells following WSP and HC treatments could be explained by the increase in GR activity, because the decrease in the GSSG concentration correlates with an increase in the GR activity. In fact, the induction of GR activity has been demonstrated with other antioxidants such as α-tocopherol, butylated hydroxyanisol, and propyl gallate (20). The induction of GR activity suggests that the critical factor was the maintenance of the low GSSG level (4).…”
