The study describes the effect of Chinese herbal medicine extracts from Scutellaria barbata on serum glycolipid hormones and pancreatic-tissue structure in type 2 diabetes mellitus (T2DM) rats. Healthy Wistar rats (n = 150) were divided randomly into two groups: healthy (30) and model (120). The healthy group was fed normally for 30d and injected citrate buffer on day 31. The model group was fed high-fat and high-sugar feed for 30d and injected STZ on day 31. On day 38 the model group was divided randomly into four groups: model group, low-, medium-, and high-dose S. barbata (n = 30 in each). 5, 10, 20, and 30dafter treatment, serum levels of FGB, INS, CORT, TG, TC, HDL-C, and LDL-C in all groups were measured and pancreatic-tissue sections were stained with hematoxylin–eosin and aldehyde fuchsine to observe the tissue structure. The results showed significantly decreased levels of FGB, INS, CORT, and LDL-C (P less than 0.01) and increased level of HDL-C (P greator than 0.05).In addition, the damaged structure of diabetic pancreatic tissue has been partially restored in the low-dose S. barbata group. These results show that low-dose S. barbata extract treatment is effective in treating T2DM.

Antioxidant status determines the susceptibility of tissues to the oxidative stress associated with diabetes and its complications; hence in the present study antioxidant status was explored in streptozotocin-induced diabetic rats treated with vanadium in the form of vanadium pentoxide nanoparticles. Vanadium pentoxide and vanadium pentoxide nanoparticles at the dose rate of 5mg/kg were administered orally in STZ (50mg/Kg) induced diabetic rats for 30 days and glimepiride (reference drug) was administered orally at the dose rate of 800 ìg/kg body weight. Vanadium pentoxide nanoparticles significantly reduced the blood glucose levels than the diabetic control and other treatment group of rats. On exploration of antioxidant status in liver, kidney and pancreas tissues, vanadium in the form of vanadium pentoxide nanoparticles outperformed the vanadium pentoxide by increasing the activities of the enzymes catalase, superoxide dismutase and glutathione peroxidase, and the concentration of reduced glutathione and by decreasing the lipid peroxide levels. The present study also showed that the restoration of antioxidant status by vanadium pentoxide nanoparticles is comparable with that of the reference drug. It can be concluded that vanadium pentoxide nanoparticles, due to its superior control over hyperglycemia and antioxidant properties, outperformed the vanadium pentoxide treatment in enhancing the antioxidant status in diabetic rats.

This study examined the changes in skeletal muscle contraction parameters after the injection of bupivacaine into the gastrocnemius muscle in diabetic rats. Forty male Wistar albino rats (230-270 g) were divided into four groups: Group I: Untreated healthy control; Group II: Healthy injected with Bupivacaine; Group III: Diabetic control; Group IV: Diabetic injected with bupivacaine. The diabetes was induced with streptozotocin in 0.1 M citrate buffer (pH 4.5) injection at 45 mg/kg in tail vein. On 7 th day of streptozotocin injection, the rats in groups II and IV were injected with 0.25% bupivacaine at 8 mg/kg body weight into the gastrocnemius muscle. Three weeks post bupivacaine application, the animals were sacrificed and right leg gastrocnemius muscle was isolated for studying various parameters such as muscle twitch, tetanic force, contraction and relaxation time and maximum contraction and relaxation rates. The blood glucose estimation showed that the rats in group III and IV developed diabetes at two days after administration of streptozotocin. The administration of bupivacaine resulted in a significant (p<0.05) increase in the muscle twitch parameters both in Group II and Group IV. Thus, bupivacaine can increase muscle contractility in diabetic muscles also.
