Antioxidative effect of BHA in soya bean lecithin‐based extender containing Glycerol or DMSO on freezing capacity of goat semen

Abstract: The aim of this study was to evaluate the effects of butylated hydroxyanisole (0 or 4 mM) along with different concentrations (5 or 7%) of glycerol (G) and dimethyl sulphoxide (DMSO) as cryoprotectant (CPAs) on freezability of goat semen. Semen was collected from four bucks (3-4 years) twice a week for five weeks. The pooled ejaculates were diluted with extender containing two different concentrations of G or DMSO in combination with BHA. Afterwards, the diluted samples were loaded into 0.25 ml straws and froz… Show more

“…Contextually, broad evidence suggests higher concentrations may not necessarily translate into better quality, but indeed may be detrimental [55]. In agreement with the present study, the use of other antioxidants such as arbutin, butylated hydroxyanisole, rosemary or lycopene, have reported a positive effect on goat sperm motility [11,44,45,56].…”

“…Based on the current results, the addition of HT at low concentrations (10 µg/mL) may improve membrane integrity, showing an increase of 14% compared to Control treatment, as reported by Hamden et al [22] when HT was added. Similarly, some authors reported an increase of membrane integrity when other antioxidants were added in extender medium [8,56,58,59].…”

The HOST was performed by incubating 30 µL semen with 300 µL of a 100 mOsm hypoosmotic solution (0.9 g fructose and 0.49 g sodium citrate dissolved in 100 mL distilled water) at 37 °C for 30 min (Rahmatzadeh et al, 2017). After incubation, 10 µL of the mixture was placed on a glass slide and covered with a cover slip to evaluate under a phase-contrast microscope (×400).…”

“…DMSO is a cryoprotectant that quickly enters sperm cells. It can be used to maintain the frozen sperm quality of bulls [115], boars [29], goats [116], and dogs [117]. In stallions, it is preferable to use DMSO or methyl formamide alone or combined with glycerol in skim milk extenders as cryoprotectants and alternatives to extenders containing glycerol only [118,119].…”
