Antiplasmodial prenylated flavanonols from Tephrosia subtriflora

Abstract: The CHCl/MeOH (1:1) extract of the aerial parts of Tephrosia subtriflora afforded a new flavanonol, named subtriflavanonol (1), along with the known flavanone spinoflavanone B, and the known flavanonols MS-II (2) and mundulinol. The structures were elucidated by the use of NMR spectroscopy and mass spectrometry. The absolute configuration of the flavanonols was determined based on quantum chemical ECD calculations. In the antiplasmodial assay, compound 2 showed the highest activity against chloroquine-sensitiv… Show more

“…purpurea, the aerial parts of T. subtriflora, and isolated flavonoids showed in vitro chemo-suppressive effect against the CQ-sensitive strains of P. falciparum (D6) [32,33]. Conversely, in this study, the ethanolic root extract of T. gracilipes was inactive (IC 50 >200 µg/mL), which could be due to…”

“…It was not cytotoxic to cancerous HepG2 cells at 100 µM while being moderately cytotoxic against non-cancerous human cells, indicating selectivity against D6 parasites [198]. The aerial parts of the related Tephrosia subtriflora were also active against P. falciparum 3D7, D6, and KSM 009 field isolate (IC 50 = 4.5–11.4 µg/mL [199]. The prenylated flavonoid MS-II ( 320 ) was subsequently isolated from the active extract and inhibited the 3D7, KSM 009, and artemisinin-sensitive F32-TEM strains.…”

“…The prenylated flavonoid MS-II ( 320 ) was subsequently isolated from the active extract and inhibited the 3D7, KSM 009, and artemisinin-sensitive F32-TEM strains. The compound was not cytotoxic against Hep2 and Vero cells at 247.5 µM, suggesting a selective antiplasmodial activity [199]. Furthermore, the root extract of Tephrosia aequilata , which inhibited 3D7 P. falciparum by 100% at 10 µg/mL, produced the new aequichalcone C ( 321 ).…”

“…Flavanonol MS-II (24) is a flavanolol with 2 (2,2-dimethylchromene) rings. In spite of having been solely tested on sensitive strains, it demonstrates a relevant variety in activity depending on the strain (see ▶ Table 3) [68]. Its selectivity toward the parasite is noteworthy, proposing that resistant strains should be tested in order to assess the compoundʼs potential.…”
