Antitumor Activity of Curcumin in Glioblastoma

Abstract: Current standard-of-care treatment for glioblastoma, the most common malignant primary central nervous system (CNS) tumor, consists of surgical resection followed by adjuvant chemotherapy and radiation (Stupp protocol), providing an overall median survival of 15 months. With additional treatment using tumor-treating fields (Optune® therapy, Novocure Ltd., Haifa, Israel), survival can be extended up to 20 months. In spite of significant progress in our understanding of the molecular pathogenesis, the prognosis … Show more

“…Curcumin (diferuloyl-methane) is a natural yellow compound isolated from the rhizome of Curcuma longa . Many reports demonstrate that curcumin shows different kinds of beneficial effects, including antioxidant [ 16 ], anti-inflammatory [ 17 ] and antitumor effects [ 18 , 19 ], while also inhibiting the p300/CBP-specific HAT activity [ 20 ]. Even though curcumin has low bioavailability, highly bioavailable forms of curcumin is expected to exhibit potential health benefits and expected to contribute to the health management [ 21 , 22 ].…”

“…Despite optimized therapy, consisting of surgery, chemotherapy, and radiation, its expected median survival remains under two years [ 1 ]. Significant progress in understanding the pathogenesis of the disease in recent decades has not been translated into an improved prognosis for patients diagnosed with GBM, which remains dismal [ 2 ]. Historically, treatment consisted of maximal surgical resection with radiotherapy alone and had a median overall survival time of approximately 12.1 months.…”

“…Notably, in OS cell lines, curcumin shows tumor-selective cytotoxicity and pro-apoptotic and anti-invasive effects [39][40][41][42][43]. Most recently, curcumin has been shown to enhance the effects of chemo-and radiotherapy on glioblastoma [44].…”
