Combined effect of montmorillonite nanoclay (MMT) (3%w/w) and Origanum majorana essential oil (MO) (0.5, 1.0, and 1.5%v/v) on microstructural (SEM, TEM, and XRD), water sensitivity, barrier, mechanical, thermal, and antibacterial properties of sodium alginate (SA) was studied. Results showed that, addition of both MMT and MO reduced water solubility of SA film up to 47% while MO increased water vapor permeability of SA/MMT films. MO improved surface hydrophobicity of SA/MMT film but tensile strength tend to decrease when both MMT and MO (≥ 1%v/v) were added into film formulation. The nanocomposite film inhibited the growth of Escherichia coli, Listeria monocytogenes, Bacillus cereus, and Staphylococcus aureus at 1.5% MO concentration confirming the functionality of the films. Finally, combined application of MMT and MO could provide a tool for developing an active SA film with minimized side effects caused by the hydrophobic nature of the essential oil. Practical application Huge amounts of synthetic packaging materials used during last decades has caused serious environmental problems resulting great efforts by both governments and researchers for finding innovative alternatives for the conventional packaging materials. Biodegradable packaging materials has been introduced as a promising solution for the problem. Biodegradable packaging materials like biopolymer based films can be produced from proteins, carbohydrate and lipids and can be replaced or complement traditional packaging systems. Up to now, biopolymer‐based packaging materials have been partially successfully used to prolong the shelf life of food products. However, there are still new developments in various fields and from various perspectives worth reporting. This study developed and characterized a biodegradable and antimicrobial nanocomposite films by incorporating MMT nanoparticles and marjoram essential oil in sodium alginate matrix.

Bovine herpesvirus 1 (BHV-1) is a highly contagious viral pathogen which causes infectious bovine rhinotracheitis in bovine worldwide. Currently, there is no antiviral prophylactic treatment available capable of the complete cure of the viral disease and facilitating recovery from latent infection in animals. The present study aimed to evaluate antiviral activities of Water Green Tea Extract (WGE) and Ethanol Propolis Extract (EPE) against BHV-1 virus comparing to commercial Acyclovir (ACV) in vitro in Madin-Darby Bovine Kidney (MDBK) cell line and in vivo in rabbits as a laboratory animal's model. The cytotoxicity assay was determined the safe dose of water green tea, and Ethanol propolis extracts and evaluated antiviral activity of each extract on infected MDBK with BHV-1. The fifteen rabbits were divided accidentally into five groups. Groups 1, 2 and 3 were inoculated with BHV-1 virus 10 7 TCID50/250 ul in nostrils and received propolis ethanol, water green tea extracts and ACV antiviral for 7 dpi respectively. Group 4 was inoculated with BHV-1 virus 107 TCID50/250 ul in nostrils without extracts or commercial drug. Group 5 was considered as control negative. Results of in-vitro study showed water green tea, and ethanol propolis extracts were potent inhibitor on BHV-1, which showed 80% protection against this virus and dropped in viral titer more than ACV. In vivo study of treated infected animals with WGE, EPE and ACV reduced clinical signs, elevated cytokines, and antibody production levels and failed re-isolated or detect DNA in blood or nasal samples swabs. Non treaded infected rabbits group developed respiratory clinical signs, humoral response and re-isolated BHV-1 and detected viral DNA of BHV-1 in blood, and nasal swabs from experimentally infected rabbits. In conclusion, propolis and green tea extracts were able to prevent virus replication and reduced CPE in MDBK cell cultures infected with BHV-1 and able to induce cytokines and antibodies levels production.

Salvadora persica L. root (Miswak) is a natural tooth cleaning tool which contains a lot of biologically active metabolites. The aim of this study is to find out the optimal method of using S. persica root as a mouth antimicrobial agent and to establish a new method for quantification of benzyl isothiocyanate (BICT) in Miswak dichloromethane extract (MDE) applying HPLC-UV. Non targeted metabolomic analysis via high performance liquid chromatography coupled with electro spray ionization tandem mass spectrometry (HPLC-PDA-ESI-MS/MS) was established for analysis of the instantly used powder (IUP) and the aqueous extract (AE). The antiviral and antibacterial activities were evaluated using MTT assay and broth micro dilution methods, respectively. HPLC-PDA-ESI-MS/MS tentatively identified 12 metabolites in IUP (glucotropaeolin, 8 flavonoid glycosides, 1 sulphated sugar derivative and 2 sugars) and 3 metabolites in the AE (2 sugars and 1 sulphated sugar derivative). IUP exhibited more potent antiviral activity against Herpes simplex virus type 1 (HSV-I) (34.79 %) than AE (25.23 %). It also demonstrated antibacterial activity against aerobic (Pseudomonas aeruginosa MIC of 6.25 mg/ml, and Bacillus sphaericus MIC 12.5 mg/ml) and anaerobic (Porphyromonas gingivalis MIC of 100 mg/ml) strains. The presence of glucotropaeolin in IUP only is suggested to be responsible for its potent antimicrobial activity in comparison to AE. A simple and validated method for quantification of BITC in MDE was established and 95 µg BITC was found to be in one gram of Miswak powder.
