This chapter reviews studies about egg-laying in the cuttlefish Sepia officinalis. Egg masses are spawned in specific mating and spawning coastal areas where mates aggregate between April and June in the English Channel and all year long in the Mediterranean Sea. Environmental cues are clearly involved in the aggregation process, but chemical communication also plays a determining role in these complex mechanisms. The successive steps of egg-laying are orchestrated by three classes of regulatory peptides: (1) neuropeptides that integrate environmental cues, (2) ovarian regulatory peptides that modulate the activity of the genital tract, and (3) sex pheromones expressed and released by the oviduct gland. After egg-laying, embryo protection is ensured for 8-10 weeks by a multilayer capsule secreted by the accessory sex glands. The oviduct gland secretes the inner layer of the egg case. The main nidamental gland secretes the main polysaccharides and glycoproteins, such as Sepia Egg Case Proteins, involved in capsule formation and in embryo protection. The accessory nidamental gland expresses specific proteins inherent in the structural organization of the gland, and hosts symbiotic bacteria. Similarly to salivary glands, this gland secretes immune factors possibly associated with gamete and/or embryo protection.

SUMMARYAn ability to sense and respond to environmental cues is essential to the survival of most marine animals. How water-borne chemical cues are detected at the molecular level and processed by molluscs is currently unknown. In this study, we cloned two genes from the marine mollusk Aplysia dactylomela which encode multi-transmembrane proteins. We have performed in situ hybridization that reveals expression and spatial distribution within the long-distance chemosensory organs, the rhinophores. This finding suggests that they could be receptors involved in binding water-borne chemicals and coupling to an intracellular signal pathway. In support of this, we found expression of a phospholipase C and an inositol trisphosphate receptor in the rhinophore sensory epithelia and possibly distributed within outer dendrites of olfactory sensory neurons. In Aplysia, mate attraction and subsequent reproduction is initiated by responding to a cocktail of water-borne protein pheromones released by animal conspecifics. We show that the rhinophore contraction in response to pheromone stimulants is significantly altered following phospholipase C inhibition. Overall, these data provide insight into the molecular components of chemosensory detection in a mollusk. An important next step will be the elucidation of how these coordinate the detection of chemical cues present in the marine environment and activation of sensory neurons.

Chemical communication among freshwater organisms is an adaptation to improve their coexistence. Here,we focus on the chemical cues secreted by the freshwater gastropod Lymnaea stagnalis, which are known to stimulate behavioural responses of Trichobilharzia ocellata (Plathelminthes, Digenea, Trematoda) miracidia. Such responses are commonly claimed to influence transmission positively, but in response to chemical cues miracidia randomly change their swimming direction. This kind of response does not necessarily increase transmission, because miracidia may be trapped at the periphery of very large snail odour-clouds, which may prevent them from approaching the snail. On the other hand, the odour clouds may be too small to improve host-localisation. To shed light on these scenarios, the spreading of molecules released around L. stagnalis (active space) was visualised by recording host-finding responses of T. ocellata miracidia when they approached snails. Behavioural responses of miracidia indicated the spreading of compounds forming an attractive active space only around the host-snail L. stagnalis, but not around sympatric non-host-snail species. The active space increased approximately linearly with the time the snail rested at the same spot and within 5 min it reached a volume of more than 30 times that of the snail. We also demonstrated in a large-scale experiment, that the active space of L. stagnalis significantly increases the transmission success of T. ocellata miracidia. Additionally, the microhabitat selection of T. ocellata miracidia was studied, demonstrating that peripheral locations near the water surface were preferred, which are also preferred sites of L. stagnalis. Improved chemoperception and microhabitat selection may have been a consequence of coevolution with snails and benefited miracidia, which became efficient transmissive stages.
