Abstract-The purpose of this study was to analyze the association between APOE alleles and serum lipemia in adolescents with low and adequate aerobic fitness. The sample was comprised of 105 boys and 151 girls (49% and 46% from rural area) of European ancestry, aged 11 to 17 years, and classified according to: 1) APOE genotype: group ε2 (ε2/3+ε2/2), ε3 (ε3/3), and ε4 (ε3/4+ε4/4); 2) aerobic fitness: adequate or low; 3) serum lipemia: elevated total cholesterol (TC), low-density lipoprotein (LDL) and triglycerides, and low high-density lipoprotein (HDL). The results showed that aerobic fitness modulates the association between APOE alleles and serum lipemia in adolescents, suggesting that adequate aerobic fitness levels exert a greater effect of reducing TC and LDL in ε2 carriers, as well as of increasing HDL and reducing triglycerides in ε3 and ε4 carriers. Os resultados demonstram que a aptidão aeróbia modula a associação dos alelos da APOE com a lipemia sérica de adolescentes, sugerindo que níveis adequados de aptidão aeróbia têm efeito maior em reduzir CT e LDL elevados nos portadores do alelo ε2, bem como o efeito maior em aumentar HDL e reduzir triglicerídeos naqueles com ε3 e ε4. Recent studies performed provide mechanistic insight into effects of the microbiota on cholesterol metabolism, but less focus was given to how cholesterol impacts the gut microbiota. In this study, ApoE−/− Sprague Dawley (SD) rats and their wild‐type counterparts (n = 12) were, respectively, allocated for two dietary condition groups (normal chow and high‐cholesterol diet). Total 16S rDNA of fecal samples were extracted and sequenced by high‐throughput sequencing to determine differences in microbiome composition. Data were collected and performed diversity analysis and phylogenetic analysis. The influence of cholesterol on gut microbiota was discussed by using cholesterol dietary treatment as exogenous cholesterol disorder factor and genetic modification as endogenous metabolic disorder factor. Relative microbial variations were compared to illustrate the causality and correlation of cholesterol and gut microbiota. It turned out comparing to genetically modified rats, exogenous cholesterol intake may play more effective role in changing gut microbiota profile, although the serum cholesterol level of genetically modified rats was even higher. Relative abundance of some representative species showed that the discrepancies due to dietary variation were more obvious, whereas some low abundance species changed because of genetic disorders. Our results partially demonstrated that gut microbiota are relatively more sensitive to dietary variation. Nevertheless, considering the important effect of bacteria in cholesterol metabolism, the influence to gut flora by “genetically caused cholesterol disorder” cannot be overlooked. Manipulation of gut microbiota might be an effective target for preventing cholesterol‐related metabolic disorders.
Apolipoprotein A5 (APOA5) and apolipoprotein E (APOE) play important roles in the metabolism of cholesterol and triglycerides. The aim of this study was to determine the allelic and genotypic distributions of the APOA5-1131T>C (rs 662799) and the APOE HhaI polymorphisms and to identify the association of both individual and combined APOA5-APOE genetic variants and the risk for dyslipidemia in children and adolescents. We genotyped 53 dyslipidemic and 77 normolipidemic individuals. The total cholesterol, triglycerides and HDL cholesterol were determined enzymatically. For APOA5 polymorphism, the presence of the allele C confers an individual risk for dyslipidemia (OR = 2.38, 95% CI = 1.15-4.89; P = 0.018). No significant differences were observed for lipid parameters among the APOA5 groups, except for a higher value of HDLc (P = 0.024) in C-carriers. The allelic and genotypic frequencies of APOE polymorphism were similar between groups and did not increase the susceptibility for dyslipidemia. None of the combined APOA5-APOE polymorphisms increased risk for dyslipidemia. We demonstrated an association between APOA5-1131T>C polymorphism and dyslipidemia in children and adolescents. This finding may be useful to guide new studies with genetic markers down a path toward a better characterization of the genetic risk factors for dyslipidemia and atherosclerotic diseases.
