Apparent Similarity of Mechanisms of Platelet Adhesion and Aggregation

Abstract: SummaryA correlation between effects of a series of compounds on platelet adhesion, aggregation, and retraction of the clot, and their effects on platelet electrophoretic mobility, ecto-ATPase activities, and AChE activity has been sought. Compounds which inhibited aggregation induced by ADP or epinephrine also inhibited platelet adhesion to glass. Six compounds which inhibited adhesion and aggregation did not inhibit clot retraction. The data suggest that the effect of thrombin on platelet electrophoretic mob… Show more

“…The high concentrations of benextramine needed to antagonize the adrenaline effect, inhibited ADPand collagen-induced aggregation. Similarly, high concentrations of phentolamine and phenoxybenzamine (100tLM) inhibit ADP-induced aggregation (Bygdeman & Johnsen, 1969), and 1 mM quinidine i inhibits collagen and ADP-induced aggregation (Mason, Read, Saba & Shermer, 1972).…”

“…Collagen-induced platelet aggregation is inhibited by phenylbutazone and sulphinpyrazone (Packham et al 1967 andMason et al 1972). On the other hand, Weily et al (1970) found no inhibition of collagen-induced platelet aggregation-by sulphinpyrazone.…”

“…Several investigations have shown that platelets adhere specifically to collagen and tissues rich in collagen, such as the basement membrane of the veins (Hugues 1962, Kjerheirn et al 1962, Zucker et al 1962, Spaet et al 1966and Baumgartner et al 1967. Testing of the aggregation of platelets in the presence of collagen has therefore become a well standardised method for assessing platelet adhesion in vitro (Bridges et al 1965a, b, Ham et al 1967, Mason et al 1972and Jobin et al 1972.…”
