Application of a lipid-coated hollow calcium phosphate nanoparticle in synergistic co-delivery of doxorubicin and paclitaxel for the treatment of human lung cancer A549 cells

Abstract: In this study, we developed a lipid-coated hollow calcium phosphate (LCP) nanoparticle for the combined application of two chemotherapeutic drugs to human lung cancer A549 cells. Hydrophilic doxorubicin (DOX) was incorporated into the hollow structure of hollow calcium phosphate (HCP), and a lipid bilayer containing hydrophobic paclitaxel (PTX) was subsequently coated on the surface of HCP. The study on combinational effects demonstrated that the combination of DOX and PTX at a mass ratio of 12:1 showed a syne… Show more

“…Following treatment of A549 cells with either the DOX-PTX-ACHN suspension or the DOX-PTX suspension, an ultrasonic grinding method was used to extract total protein from A549 cells. The concentration of total protein obtained was determined using the BCA method [ 26 ]. A solution of the protein samples and buffer at a ratio of 4 : 1 was boiled for 5 min.…”

“…36,37 As a result, DOX is often used together with other agents to further improve its anticancer performance as well as reduce the side effects. 38,39 Co-delivering systems have been widely explored by precious works, in which polymer-based DDS, such as HA, 40 polyethylene glycol, 4 chondroitin sulfate, 36 etc have shown great promise in overcoming the abovementioned dilemma. However, compared with previous works' focus on co-delivering QC and DOX, 41,42 persistent work is still required on the fabrication of a reliable and reproducible hybrid platform with decent drug-loading capability as well as tumor targetability to enhance the co-delivering efficacy in GC chemotherapy.…”

“…For instance, the LCP nanoparticle has been used to deliver gene therapy agents such as siRNA and expression vectors (55,56). It has also been used to deliver many therapeutical chemicals such as gemcitabine triphosphate (20,21), doxorubicin and paclitaxel (57). Due the similarity between A*TPγS and gemcitabine triphosphate, we choosed to use this nanoparticle for intracellular A*TPγS delivery in this study.…”
