Aquaglyceroporins: Drug Targets for Metabolic Diseases?

Abstract: Aquaporins (AQPs) are a family of transmembrane channel proteins facilitating the transport of water, small solutes, and gasses across biological membranes. AQPs are expressed in all tissues and ensure multiple roles under normal and pathophysiological conditions. Aquaglyceroporins are a subfamily of AQPs permeable to glycerol in addition to water and participate thereby to energy metabolism. This review focalizes on the present knowledge of the expression, regulation and physiological roles of AQPs in adipose… Show more

“…Obesity is regularly associated with hormonal dysregulation [ 41 , 42 ]. In addition, obesity is related with alterations in the expression and function of the aquaglyceroporins and consequently in glycerol transport [ 43 , 44 ]. Obese men are known to have a positive correlation with abnormal sperm morphology, decreased sperm concentration and sperm motility, decreased serum testosterone and increased E2 levels compared with healthy men, which consequently leads to compromised fertility [ 45 , 46 , 47 ].…”

“…AQPs 11 and 12 are more distantly related to other mammalian AQPs, based on amino acid sequence; functions and regulation remain to be fully defined [ 19 ]. Diverse contributions of AQP classes could augment pro-cancer conditions by enhancing signaling via nitric oxides or hydrogen peroxide [ 20 , 21 , 22 , 23 ], facilitating gas exchange of O 2 and CO 2 [ 24 , 25 ], promoting cell cycle progression [ 26 ], providing metabolic support [ 27 , 28 ], mediating dual ion and water fluxes for localized control of process extension [ 12 ], driving angiogenesis [ 29 ], boosting mechanical compliance needed for rapid volume changes [ 6 , 30 ] and other processes. Dynamic translocation between membrane and intracellular vesicle pools is controlled by signaling; for example, AQP2 localization depends on cyclic AMP-dependent kinase activity [ 31 ], and AQPs are associated via protein–protein interactions into signaling complexes [ 32 , 33 , 34 ].…”

“…MR309, a Sigma-1 antagonist is currently in Phase II clinical trial for the treatment of neuropathic pain) [31,49,50]. On the other hand, aquaporins are still largely unexplored from a medicinal chemistry standpoint and have been recognized as druggable molecular targets only recently [17,[51][52][53][54][55]. To the best of our knowledge, this represents the first report involving modulators of both AQP and S1R.…”
