Arabidopsis ADF5 promotes stomatal closure by regulating actin cytoskeleton remodeling in response to ABA and drought stress

Abstract: ADF5 promotes stomatal closure by regulating actin filament dynamics, and members of the ABF/AREB transcription factor family may serve as potential upstream regulators of ADF5 in the drought stress/ABA signaling pathway.

“…Naturally, the eight proteins induced in response to drought in all genotypes (DHAR1, KAT, LTP1, HSP 18.6, GRXC6, RNS3, ADF3, and TIM18) are prime candidates for the selection of drought-tolerance markers. Most of these proteins have been reported to play important roles in response to plant hormones [14][15][16][17] and are involved in detoxification of reactive oxygen species (ROS) [15,18,19]. Protein transport to mitochondria through the membrane is facilitated by TIM complexes, potentially delivering antioxidant enzymes to mitochondria and enabling ROS detoxification during water deficits.…”

“…Its role in the leaves of droughted plants remains unknown. While ADF-3 and other members of the ADF family, such as ADF-5 [16], have a function in abscisic acid-induced stomatal closure and therefore transpiration, it needs to be established that stomatal complexes alone could strongly influence the abundance of this group of proteins.…”

“…It was shown that Atabf4/areb2 and Atabf3 mutants were more sensitive to drought than WT, whereas overexpression of AtABF2/AREB1 ensured better drought tolerance due to increased expression of LEA genes ( Fujita et al, 2005 ; Yoshida et al, 2010 ). AtABFs/AREBs promote stomatal closure and chlorophyll catabolism ( Gao et al, 2016 ; Qian et al, 2019 ). Importantly, AtABF2/AREB1, AtABF4/AREB2, AtABF3, and AtABF1 show redundancy during regulation of ABA-mediated drought responses.…”

“…We previously showed that both knockout/knockdown and overexpression of the F-actin reorganization regulators SCAB1 and CKL2 delayed stomatal closure (32,35). A defect in ADF5 also resulted in delayed stomatal closure (51), while overexpressing ADF1 led to partially or entirely closed stomata (52). The morphological changes in the guard cell vacuole are closely related to stomatal movement (53).…”
