Arsenic Excretion after Treatment of Arsenic Poisoning with DMSA or DMPS in Mice

Abstract: Abstract-The effects of 2,3-dimercaptosuccinic acid (DMSA) and 2,3-dimercap topropane-1 -sulfonic acid, Na salt (DMPS) on arsenic excretion in arsenic poisoning were studied using ICR mice. One group of mice was given arsenic trioxide (5 mg As/kg, s.c.) and another two groups were given DMSA or DMPS (100 mg/kg, i.p.) immediately after administration of the arsenic (5 mg/kg, s.c.). Arsenic excretion in urine and feces was determined by atomic absorption spectro photometry.

“…The pellets obtained were re-suspended in 1 ml PBS and used for measurement of platinum and DNA contents. Measurement of platinum was carried out according to the method described previously (19,20). Briefly, 1 ml 50% Mg(NO 3 ) 2 was added to the pellet followed by heating for 30 min at 70°C.…”

“…Excretion of arsenate and arsenite following parenteral exposure of animals is similar to that seen following oral exposure. In rabbits and mice, urinary excretion within 8 hours usually accounts for about 50-80% of the dose (Maehashi and Murata 1986;Maiorino and Aposhian 1985;Vahter and Marafante 1983). Somewhat lower levels (30-40%) are excreted in the urine of marmoset monkeys (Vahter and Marafante 1985;Vahter et al 1982), probably because of the absence of methylation in this species.…”
