Artificial fertilization and induction of triploidy and meiogynogenesis in the European sea bass, Dicentrarchus labrax L.

Abstract: The European sea bass, Dicentrarchus labrax L., was successfully subjected to chromosome manipulation. Triploidy was induced by cold-shocking e gs at 0-2°C for 20 min starting 5 min after fertilization in order to prevent the extrusion of the secon%polar body. Meiogynogenesis was also obtained by fertilizing eggs with W-irradiated sperm (3300 and 6600 erg m-) and subsequently doublin the chromosome set by meiotic block as above. The commercial advantages of culturing triploicf and gynogenetic sea bass are disc… Show more

“…Arti¢cial fertilization was carried out within 10 min from gamete collection, mixing 1 part of the eggs, 4 parts of seawater and 1 part of sperm diluted 1:10 with an extender solution (375 mosmoles kg À1 KCl, pH 7.35) as reported by Colombo, Barbaro, Libertini, Benedetti, Francescon and Lombardo (1995). A full stripping of eggs was performed in all the females and, in case of good egg quality, sperm was immediately collected from the males.…”

“…Although spontaneous reproduction occurs easily in captive seabass breeders (Devauchelle & Coves, 1988), and the preliminary results on sperm characterization and management (Billard, 1984) have been exploited to establish artificial insemination routines (Zanuy et al, 1994;Felip et al, 1997), artificial insemination is still unpredictable (Colombo et al, 1995). The present development of seabass aquaculture and the increase in genetic studies in this species, require reliable artificial insemination and gamete management protocols including sperm economy and storage since males do not provide large volumes of semen (Zohar et al, 1984).…”
