Asp-698 and Asp-811 of the Integrin α4-Subunit Are Critical for the Formation of a Functional Heterodimer

Abstract: The amino acid motif LDV is the principal binding site for ␣ 4 integrins in fibronectin, and homologous motifs are recognized in vascular cell adhesion molecule-1 and MAdCAM-1. Three conserved LDV motifs (LDV-1 to 3) occur in the ectodomain of the human and mouse ␣ 4 -subunit, the functions of which are unknown. “…Since it has been shown that ␣1␤1 and ␣2␤1 integrins are essential for the collagen-dependent stimulation of the expression of collagenase 3 mRNA in skin fibroblasts (25), it can be assumed that integrin-type collagen receptors, including ␣1␤1, ␣2␤1, ␣10␤1, and ␣11␤1 integrins, are involved in the collagen-dependent stimulation of collagenase 3 expression in synovial fibroblasts (40)(41)(42)(43) Furthermore, the involvement of the integrinbinding motifs RGD and LDV in the regulation of collagenase 3 expression in synovial fibroblasts was studied. These integrin-binding motifs have been shown to be functional in several extracellular matrix molecules, including fibronectin and vitronectin, as well as in degraded collagen (37,44,45).
