ASP53, a thermostable protein from Acacia erioloba seeds that protects target proteins against thermal denaturation

Abstract: ASP53, a 53 kDa heat soluble protein, was identified as the most abundant protein in the mature seeds of Acacia erioloba E.Mey. Immunocytochemistry showed that ASP53 was present in the vacuoles and cell walls of the axes and cotyledons of mature seeds and disappeared coincident with loss of desiccation tolerance. The sequence of the ASP53 transcript was determined and found to be homologous to the double cupin domain-containing vicilin class of seed storage proteins. Mature seeds survived heating to 60°C and t… Show more

“…In contrast to the seed developmental stage, many transcription factors (MYB, AP2, and NAC), and gene encoding methyltransferase and histone, are differentially expressed ( Tian et al., 2016 ). And the DT relative genes such as LEA and HSP are down-regulated suggesting the loss of these proteins alters the DT of a germinating seedling ( Mtwisha et al., 2007 ; Ginsawaeng et al., 2021 ).…”

“…A proteinaceous α‐amylase inhibitor CL‐AI contains one α‐chain and one β‐chain possessing the same conserved cupin domain that could inhibit various α‐amylases and delay the seed germination of wheat, rice, and maize as well as the growth and development of potato beetle larva (Wang et al ., 2018b). The Acacia erioloba ASP53 as homologous to the cupin domain containing vicilin class of seed storage proteins that protects target proteins against thermal denaturation (Mtwisha et al ., 2007). In rice, the lack of cupin domain containing globulin synthesis during seed development alters the accumulation of seed storage proteins with the reductions in glutelin while prolamin level conserved in rice (Lee et al ., 2015).…”
